氢化可的松油水分配系数及其皮肤渗透速率的相关性研究

摘  要:目的探讨氢化可的松的辛醇/水分配系数与其皮肤渗透速率之间的相关性。方法采用高效液相色谱法测定氢化可的松(HC)在不同pH环境中的辛醇/水分配系数,以及HC在不同pH环境中的皮肤渗透速率,并考察两者之间的相关性。结果在研究的pH范围内,HC的辛醇/水分配系数与其经皮渗透速率的回归方程为y=0.795x+25.316,r=0.985。结论在pH为4~10的范围内,氢化可的松的辛醇/水分配系数与经皮渗透速率之间有较好的线性关系,初步提示通过测定药物的辛醇/水分配系数可以预测其透皮吸收性能。Objective To investigate the correlation between the oil/water partition coefficient(PC) and transdermal penetrating rate (PR) of hydrocortisone. Methods Based on the pH partition principle, different PC and PR of hydrocortisone were obtained and determined by HPLC method. The correlation between PC and PR was studied. Results The correlation between PC and PR followed a linear relationship, whose equation was y = 0. 795x + 25. 316, r = 0. 985. Conclusions Within the range from pH 4 to 10, there was a good linear relationship between PC and PR of hydrocortisone, which initially indicated that the transdermal absorpting ability of the drug could be predicted by detecting its PC.
