摘  要:建立血液与唾液中葛根素的HPLC分析方法.色谱柱为ODS C18柱(150 mm×4.6 mm,大连依立特公司);流动相为乙腈–含0.8%冰醋酸的水溶液(22∶78,V/V);流速为0.8 mL/min;柱温为30℃;检测波长为250 nm.葛根素血浆和唾液质量浓度标准曲线范围分别为5.0~350.0μg/mL和0.1~5.0μg/mL,该方法血液与唾液最低定量限分别为5.0μg/mL和0.1μg/mL,平均回收率为97.5%~103.0%和86.4%~108.0%,高、中、低三个质量浓度血浆和唾液标准样品的日内精密度RSD在3.3%~6.4%和2.7%~3.9%,日间精密度RSD在1.6%~3.6%和3.5%~4.6%.本方法同时适用于血液及唾液样品中微量葛根素的高效液相色谱分析,取样量小,灵敏度高,操作简便易行,为葛根素的临床药动学研究提供了方法学基础.To establish determination of puerarin in plasma and saliva samples by HPLC. Chromatography column was ODS C18 Column (250 mm×4.6mm, Yilite company in Dalian) ; Mobile phase was aeetonitrile - water(containing 0.8% glacial acetic acid , 15: 85, V/V) ; the flow - rate was 0.8 mL/min;temperature at 30℃ and detection wave length at 250 nm. In plasma and saliva, standard curve of Puerarin ranged with 5.0 - - 350.0 μg/ mL and 0.1 - 5.0 μg/mL respectively, the LOQ was 5.0 μg/mL and 0.1μg/mL, the average recovery was 97.5%- 103.0% and 86.4% - 108.0% , the RSD of intra - day was 3. 3% -6.4% and 2.7% - 3.9% , the RSD of inter - day was 1.6% - 3.6and 3.5% -4. 6%. The high performance liquid chromatographic (HPLC) method could be used simultaneously for determination of puerarin in plasma and saliva samples, which is sensitive and simple for small - biological samples analysis. It was suggested that the methodology study may be used in clinical pharmaeokineties of puerarin.
