摘 要:为考察不同温度、不同冷冻保护剂对好氧颗粒污泥(aerobic granularsludge,AGS)储存的影响,试验采用SBR反应器培养AGS,在AGS成熟阶段将其利用不同方法储存:R1(室温,蒸馏水)、R2(4℃,蒸馏水)、R3(4℃,10%(V/V)海藻糖溶液)、R4(-20℃,10%(V/V)海藻糖溶液)、R5(4℃,10%(V/V)蔗糖溶液)、R6(-20℃,10%(V/V)蔗糖溶液).储存5周后,利用方法R1、R2、R3、R5保存的AGS由浅黄色变为褐色,并维持原有形态;按照方法R4、R6保存的AGS出现了少量破碎,颜色保持淡黄色.将这6种方法保存的AGS进行活性恢复,13个周期后用各种方法储存的AGS对COD去除率均达到85%以上;SOUR在AGS恢复过程中均呈上升趋势,各种方法储存的AGS的EPS在储存后都有所增加;AGS恢复后,EPS又基本恢复到储存前的水平.本试验表明:AGS具有较强的应对外界不良条件的能力,并且能在恢复后维持颗粒形态和活性,其中用4℃、蒸馏水储存AGS的效果最好;冷冻保护剂对AGS的储存没有起到明显的作用,这是由于AGS的结构和传质特点所致.ix preserved methods were used to investigate the characteristics of storage and re-activation of aerobic granular sludge(AGS),the methods included R1(room temperature,distilled water),R2(4℃,distilled water),R3(4℃,10%(V/V)fucose),R4(-20℃,10%(V/V)fucose),R5(4℃,10%(V/V)sucrose)and R6(-20℃,10%(V/V)sucrose).Five weeks later,color of the AGS preserved by method R1,R2,R3 and R5 turned from light yellow to brown,the physical characteristics and structure stability of the AGS were found stable;a few AGS preserved by method R4 and R6 broke up,maintaining the yellow color.From cycle 13 onwards,the COD removal efficiencies of the AGS preserved by all the methods were higher than 85%.During the re-activation,SOUR showed an upward trend.After storage,the extracellular polymeric substances(EPS)of the AGS with different preserved method all increased.After re-activation,EPS all returned to the level before storage.This study shows that AGS had a strong resistance to the external adverse conditions,maintaining the physical characteristics and structure stability.The best preserved method was 4℃,distilled water.It shows that temperature played an important role on the preservation of AGS.Due to the physical characteristics,structure and substrate diffusion of AGS,cryoprotectants did not play a significant role.
