摘  要:【目的】建立吉西他滨(GEM)的高效液相色谱(HPLC)检测方法,探索微透析采样技术实现吉西他滨浓度动态测定的可行性。【方法】对所建立的HPLC检测方法的色谱条件进行方法学考察;采用浓差法(增量法、减量法)测定吉西他滨的体外回收率,考察灌流液流速、药物浓度等对回收率的影响以及体外回收率的稳定性。【结果】所建立的HPLC检测方法在实验研究所要求的浓度范围内,线性关系良好,峰形理想,精密度及稳定性均满足要求;增量法及减量法分别测得的回收率与释放率一致;回收率的大小与吉西他滨浓度无关。【结论】微透析采样技术可用于吉西他滨的浓度动态测定,体内研究中可利用反透析法测定吉西他滨的探针回收率。Objective To establish a HPLC method for the measurement of gemcitabine concentration in vitro,and to explore the feasibility of measuring gemcitabine concentration dynamically by microdialysis technology.Methods The chromatographic conditions of the established HPLC method were investgated.In-vitro recovery of gemcitabine was detected by concentration difference method(increment method and decrement method),and the stability of in-vitro recovery was observed.The influences of the flow rate and concentrations of the perfusate on gemcitabine recovery were investigated.Results The established HPLC presented good linearity with satisfactory precision and stability in the range of required concentrations.Recoveries and release rates detected by increment methods were similar to those detected by decrement methods.The concentration of gemcitabine showed no effect on recoveries.Conclusion The microdialysis technology can be applied in dynamic measurement of gemcitabine in vitro.Gemcitabine concentrations in the microdialysis samples can be determined by HPLC.The results indicate the feasibility of microdialysis technology in dynamic detection of gemcitabine in vivo.
