摘  要:为探索日本血吸虫(中国大陆株)22.6kDa 抗原(Sj22.6)编码基因用作核酸疫苗的可行性,将pCMV/Sj22.6基因重组质粒经肌肉注射免疫了一批BALB/c小鼠并进行攻击感染试验,结果表明此重组质粒能在小鼠体内持续存在、稳定表达并诱导小鼠产生特异性的抗Sj22.6 抗体。The gene fragment of 22 6kDa antigen of Schistosoma japonicum was reconstructed by PCR and subcloned into eukaryotic plasmid pCMV β.The pCMV/Sj22 6 recombinant plasmid was used for BALB/c mice vaccination/challenge experiment.The pCMV/Sj22 6 plasmid is proved by PCR to be still in quadriceps,heart and diaphragm muscle cells by week 16 and can express Sj22 6 protein in quadriceps cells,which has the same physiochemical nature as the native antigen and recombinant protein expressed in E coli,proved by Western blot.Also,with soluble adult worm antigens and rSj22 6 antigen,anti Sj22 6 specific IgG antibodies in pooled immune sera could be detected by Western blotting,and its level was determined by ELISA.However,worm recovery and liver tissue egg counts showed that the recombinant plasmid failed to induce a protection against the challenge with S japonicum cercariae.But according to the protections induced by Sj22 6 protein vaccines and its gene expression in muscle cells showed above,it is still hopeful that.The protection in mice with pCMV/Sj22 6 DNA vaccine could be induce by reforming some approaches.
