摘  要:以2-型猪链球菌(SS2)保护性抗原RfeA推定的氨基酸序列为基础,采用Kyte-Doolittle法分析蛋白的亲水性,Emini法预测蛋白的表面可能性,以及Jameson-Wolf法预测蛋白的抗原指数;辅以Garnier-Robson法、Chou-Fasman法和Karplus-Schulz法对蛋白二级结构中柔性区域的分析,预测SS2保护性抗原RfeA的B细胞表位。结果表明,推测RfeA的B细胞表位位于RfeA蛋白N-端第21~27、53-60、104~117、140~160区域。用多参数预测SS2保护性抗原RfeA的B细胞表位,为实验研究SS2的多表位疫苗奠定基础。The secondary structure flexible region of protective antigen RfeA against Streptococcus suis serotype 2(S.suis 2)was analyzed by the Garnier-Robson's method,the Chou-Fasman's method and the Karplus-Schulz's method based on their putative amino sequence.Hydrophilicity plot,surface probability plot and antigenicity index were obtained by the methods of Kyte-Doolittle,Emini and Jameson-Wolf,respectively.B cell epitopes for RfeA were predicted based on these results The predicted B cell epitopes for RfeA were located at the N-terminal No.21~27,53-60,104~117,140~160.Taken together,prediction of the B cell epitopes for protective antigen RfeA against S.suis 2 based on multiple parameters would lay foundation for experimental research of multi-epitope vaccine against S.suis 2.
