缩泉丸对肾虚多尿大鼠垂体ACTH mRNA及蛋白表达的影响

摘　　要：目的通过研究缩泉丸对肾虚多尿大鼠垂体ACTH mRNA及蛋白表达的影响,阐明其补肾缩尿的机制。方法采用腺嘌呤造成大鼠肾虚多尿模型,利用酶联免疫分析法分别检测正常对照组、模型对照组、缩泉丸(高、中、低剂量)组、金锁固精丸组动物血中ACTH、ALD的含量;并采用RT-PCR、免疫组化的方法检测各组动物垂体ACTH mRNA及蛋白的表达。结果与模型对照组比较,缩泉丸中、高剂量组可显著提高肾虚多尿大鼠血中ACTH、ALD含量(P<0.05～0.01);缩泉丸高剂量组可使肾虚多尿大鼠垂体ACTH mRNA表达明显增高(P<0.01);缩泉丸低中高剂量组均可使肾虚多尿大鼠垂体ACTH蛋白阳性产物的表达增强,表达区域广泛分布于前叶和中间叶。结论缩泉丸可上调肾虚多尿大鼠垂体ACTH mRNA及蛋白的表达,增强ACTH的功能,从而促进ALD的合成,发挥调节水液代谢的作用。Objective To explore the mechanism of Suoquan Capsules（SC） in nourishing kidney and reducing urine by discussing the influence SC on mRNA and protein expression of adrenocorticotrophic hormone （ACTH） in hyperdiuresis rats with kidney deficiency. Methods We applied adenine to establish the rat model of diuresis with kidney deficiency, detected the contents of ACTH, aldosterone（ALD） in blood , examined mRNA and protein expression of pituitary ACTH with enzyme linked-immunosorbent assay （ELISA） and RT-PCR in normal group, model group, high-, middle- and low-dosage SC groups, and Jinsuo Gujin Pills group. Results Compared with the model group , high- and middle-dosage SC could remarkably increase the contents of ACTH and ALD in the blood of model rats （P 〈 0.05 or P 〈 0.01）, high-dosage SC increased ACTH mRNA expression（P 〈 0.01 ）, and high-, middle- and low- dosage SC enhanced the protein expression of ACTH extensively in the anterior lobe and intermediate lobe of the rat brain. Conclusion SC can regulate the body fluid metabolism through promoting the formation of ALD by up-regulating ACTH mRNA and protein expressions in diuresis rats with kidney deficiency.
