Objective To investigate the protective effect of total timosaponin (TT) on chronic mildly stressed (CMS) mice and to explore its possible therapeutic mechanism. Methods The sucrose solution consumption test was used to observe the antidepressant effect of TT, and step-through test was used for the observation of learning and memory of CMS mice. Plasma levels of adrenocorticotropic hormone (ATCH) and cortisol were detected with radioimmunoassay, and brain-derived neurotrophic factor(BDNF) in the hippocampus was examined with enzyme-linked immunosorbent assay. Results Treatment with TT(25 and 50 mg·kg-1) markedly enhanced the consumption of sucrose solution in CMS models. Administration of TT(50 mg·kg-1) significantly increased escaping latency and decreased the number of errors. Biochemical studies showed that the increase of ATCH and cortisol, and the decrease of BDNF in CMS mice were significantly ameliorated by TT. Conclusion TT could improve the depression state and promote learning and memory impairment in CMS mice. The mechanism may be related with the suppression of the hyperfunction of hypothalamic-pituitary-adrenal axis and with the increase of BDNF level in the hippocampus.
