细菌亚单位疫苗免疫小鼠的细胞免疫指标检测

摘　　要：将嗜水气单胞菌的胞外毒素(HEC) 和胞外蛋白酶(ECPase) 与迟缓爱德华氏菌脂多糖(LPS)、脱毒多糖(PS) 偶联, 制成3 种亚单位偶联疫苗 (HEC-PS、HEC-LPS、ECPase-PS), 分别免疫小鼠。另设这两种细菌的全菌灭活苗免疫及注射生理盐水对照组, 每组8 只。在第2 次免疫后第7 天用四甲基偶氮唑盐比色法 (MTT), 检测脾淋巴细胞转化率与腹腔巨噬细胞吞噬活性, 并检测各组小鼠的全菌凝集抗体。结果表明, 亚单位疫苗的这两项细胞免疫指标明显高于对照组,但略低于全菌灭活苗免疫组, 而细胞免疫指标与抗体水平无对应的平行关系。With the extracellular proteins (HEC toxin or ECPase)of Aeromonas hydrophila (Ah)and the lipopolysaccride(LPS)or polysaccride(PS)of Edwardsiela tarda (Et),three conjugated subunit vaccines(CSV)were prepared,designated as HEC PS、HEC LPS and ECPase PS.48 mouses divided as 6 groups were immunized with the above three CSV,killed whole bacteria vaccine (WCV)Ah or Et and PBS separately.At the 7th day after the second immunization,the stimulation index(SI)of the spleen lymphocyte transformation and the A 570 of abdominal cavity macrophagic activity (AMA)were measured with MTT method.Meanwhile the whole bacteria agglutination titer was also tested.The result showed that as far as the value of the SI and AMA of the mouses,WCV was the best,CSV was better than the control mouses.No relationship was found between the SI/AMA and the titer of whole bacteria agglutinating antibody.
