斑鳜肌球蛋白轻链1基因cDNA的克隆及其分析

摘　　要：为提取斑鳜肌肉组织的总RNA,应用PCR法扩增肌球蛋白轻链1基因(MLC1)。结果获得斑鳜MLC1(基因库登录序列号为:GQ282999),cDNA序列的全长为579 bp,编码192个氨基酸,理论相对分子质量为20 778.57,等电点为4.51,该序列具有2个EF-手相结构。氨基酸序列比对结果表明,第2个EF-手相结构中的Ca2+结合域非常保守,氨基酸序列同源性为100%。The total RNA was isolated from the skeletal white muscle of Siniperca scherzeri. The myosin light chain 1 （MCL1） was amplified using PCR technology,and then was cloned. The length of the myosin light chain 1 （MCL1） cDNA is 579 bp,and encodes 192 amino acids ,which includes two potential EF-hand structures. The putative protein of MLC1 shows predicted molecular weights of S0 778.57 with a theoretical pI of 4.51. Compared with the MLC1 of others reported,the homology of the second potential EF-hand structure of the potential Ca^2＋ binding domain was well conserved among the MLCls of all species.
