珍菊降压片中西药物配伍对高血压相关基因表达的影响

摘　　要：目的采用血清药理学方法,研究珍菊降压片及组方中中药部分与西药部分对ECV-304人脐静脉内皮细胞可能与高血压相关的候选基因表达变化的影响。方法将ICR小鼠随机分成4组:空白对照组、复方组(280 mg/kg)、相应剂量中药组(274 mg/kg)、相应剂量西药组(6.2 mg/kg),每组20只。连续给药7d,末次给药后2 h,眼眶取血,分离含药血清。选取ECV-304人脐静脉内皮细胞1传4,24 h后,将过滤后的血清加入含10%胎牛血清的5 mL培养基中,于37℃孵育箱培养,24 h后观测细胞表型、细胞周期变化,并抽提ECV-304细胞RNA,反转录,Cy3标记和Cy5标记,作Affimatrix表达谱芯片分析。结果各组含药血清对ECV-304细胞无明显毒性作用,但可促进其增殖。各组有着相同的和不同的可能与高血压相关的候选基因表达变化,复方组表现出不同于中药组、西药组的作用靶点。结论珍菊降压片中西药物配伍后可以发挥出不同于使用中药或化学药物单独使用时的作用,二者具有协同作用。Objective To study the effect of Zhenjujiangya tablets,traditional chinese medicine part and chemical drug part of the prescription on the on the expression of candidate genes related to hypertension in ECV-304 human umbilical vein endothelial cells HUVEC.Methods Eighty ICR mice were randomLy divided into four groups,blank control group,the zhenjujiangya tablet group（280 mg/kg）,traditional chinese medicine group（274 mg/kg）,and chemical drug group（6.2 mg/kg）,with twenty mice in each group.Blood was collected from the mice after p.o.administration for seven days.Serum containing active ingredients were separated and used to incubate ECV-304 HUVEC for 24 hours to obtain HUVEC at various cell cycles.Flow cytometer was used to measure the changes in cell cycle of HUVEC.The RNA was extracted and used to measure affimatrix expression spectrum by gene chip.Results The serum showed no significant toxic effect for HUVEC and could promote the proliferation of HUVEC.The changes of gene expression induced by three different treatments have similarities and differences.Compared with the changes led by traditional Chinese medicine and chemical drug group,Zhenju Jiangya tablets group showed different gene targets.Conclusion The effect of Zhenjujiangya tablets,which contains both traditional Chinese medicines and chemical drugs are developed not only by the two groups separately,but also by the synergistic effect of two groups.
