摘  要:目的阐明胰岛素(In)与硒(Se)联合作用对糖尿病(DM)大鼠糖脂代谢及对骨骼肌细胞GLUT4表达的影响。方法腹腔注射链脲佐菌素(STZ)50 mg/kg复制大鼠DM模型,单用及联合应用胰岛素1 U/kg.d和硒180μg/kg.d4周后,分别检测血糖(BG)、糖化血红蛋白(HbA1c)、血脂(BL)的变化以及骨骼肌细胞膜上GLUT4表达的变化。结果联合用药组(DM+In+Se)的血糖和HbA1c较单用胰岛素组(DM+In)和单用硒组(DM+Se)明显下降(P<0.01);DM+In+Se的TC和TG较DM+In组和DM+Se组明显下降(P<0.01)。Western blotting和免疫组化结果均显示:胰岛素和硒联合应用能明显增加DM大鼠骨骼肌细胞膜上GLUT4的表达(P<0.01)。结论 DM+In+Se降低BG、HbA1c和BL的作用明显优于单用相同剂量的DM+In或DM+Se;DM+In+Se与增加肌组织细胞膜GLUT4蛋白的表达量而增强胰岛素信号转导,纠正糖脂代谢紊乱有关。Objective To explore the effect of insulin and selenium in combination on blood glucose and blood lipid metabolism and glucose transporter 4(GLUT4) level in skeletal muscle of streptozotocin(STZ)-induced diabetic rats.Methods Diabetic animal models were established by intraperitoneal injection of STZ into the rats.The diabetic rats were treated with insulin [1U/(kg·day)],selenium [180μg/kg·day)],and insulin combined with selenium for 4 weeks.The levels of blood glucose,hemoglobin A1c and blood lipid were determined and the level of GLUT4 expression in skeletal muscle was examined.Results Insulin in combination with selenium could significantly lower blood glucose,HbA1c and blood lipid levels(P〈0.01).Western blotting and immunohistological results showed that insulin in combination with selenium could significantly restore the disturbance in GLUT4 levels in skeletal muscle(P〈0.01).Conclusion There is a synergetic effect between insulin and selenium,and treatment of diabetic rats with combined doses of insulin and selenium is effective in normalizing blood glucose and blood lipid,and correcting altered GLUT4 distribution in skeletal muscle of diabetic rats.The two in combination can increase insulin signal transduction by upregulating GLUT4 in skeletal muscle,eventually improving glucose and lipid metabolism disorder.
