滋阴平肝复方对阴虚阳亢型高血压大鼠血管钙化及血压影响的实验研究

摘　　要：目的通过建立阴虚阳亢型高血压大鼠的动物模型,系统观察滋阴平肝复方对阴虚阳亢型高血压大鼠血压及血管硬化的调控作用。方法通过灌服附子汤的方法建立阴虚阳亢型高血压的实验动物模型,通过采用滋阴平肝复方高、中、低剂量干预,采用尾套法测定血压变化,采用荧光显微技术检测肌细胞中钙含量变化;观察不同剂量干预下,各组大鼠尾动脉血压及主动脉平滑肌细胞中钙含量变化情况。结果用药前各组大鼠尾动脉血压无明显差异,采用不同干预方法后,高、中、低剂量中药干预组及西药组大鼠尾动脉血压均有明显下降(P<0.05),其中高剂量组血压下降幅度较西药组大〔(101.19±8.33)mmHg VS(107.26±10.33)mmHg,P<0.05〕,而对照组无明显变化(P>0.05);高、中、低剂量中药干预组及西药组大鼠主动脉平滑肌细胞中钙含量均有不同程度下降(P<0.05),其中高、中剂量组钙含量下降幅度较西药组大〔(172.74±40.17)nmol/L,(221.79±34.17)nmol/L VS(366.41±39.53)nmol/L,P<0.05〕,低剂量组与西药组对比无显著差异(P>0.05)。结论 滋阴平肝法复方对阴虚阳亢型高血压大鼠血压具有较好的调节作用,能有效拮抗其血管钙化过程,高、中剂量组疗效优于西药。Objective To systematic observating the regulate and control effection of Ziying Pinggan methods acting on blood pressure and angiosclerosis by modeling rat with type of deficiency of YIN leading hypertension.Methods Modeling experimental rat with type of deficiency of YIN leading hypertension by pouring orally monkshood decoction and interventing measure implemented by high,moderate,low dose of Ziying Pinggan Chinese Traditional Medicine recipe respectively.The change of blood pressure were monitored by tail-cuff,and calcium content in myocyte monitored by fluorescence microscopy.Observing the change of caudal arterial blood pressure and calcium content in smooth muscle myocyte of aorta in groups of different dose of Ziying Pinggan Chinese Traditional Medicine recipe.Results There had no significantly deference among the groups of rat model with type of deficiency of YIN leading hypertension before intervening（P〈0.05）,but had significantly decreasing in blood pressure among the groups of high,moderate,low dose of Ziying Pinggan Chinese Traditional Medicine recipe and Western medicine after intervening（P〈0.05）.The descender of blood pressure in high dose group was significantly greater than that of Western medicine group（（101.19±8.33）mmHg VS（107.26±10.33） mmHg,P〈0.05）,but had no significantly change in control group（P〈0.05）.There had significantly decrease in calcium content in smooth muscle myocyte of aorta in groups of different dose of Ziying Pinggan Chinese Traditional Medicine recipe and Western medicine group（P〈0.05）.In all of them,the descender in groups of high and moderate dose were significantily greater than that of Western medicine group（（172.74±40.17）nmol/L,（221.79±34.17） nmol/L VS（366.41±39.53） nmol/L,P〈0.05）,but had no significantily defrence betwween low dose group and Western medicine group（P〈0.05）.Conclusion Ziying Pinggan Chinese Traditional Medicine recipe has good blood pressure control effection on type of deficiency of YIN l
