龟鹿二仙胶汤及其拆方对大鼠软骨细胞凋亡基因表达的影响

摘　　要：目的:通过观察龟鹿二仙胶汤及其拆方对SD大鼠软骨细胞凋亡基因表达的影响,比较龟鹿二仙胶汤中各药物组成在全方中的作用。方法:取1月龄SD大鼠膝关节软骨建立软骨细胞体外培养体系,采用实时荧光定量PCR检测Bax、Bcl-2、Caspase-3、p53mRNA表达。结果:龟鹿二仙胶、龟甲、鹿角、人参、枸杞、盐酸氨基葡萄糖均可显著抑制大鼠软骨细胞Bax、Caspase-3、P53的表达;龟鹿二仙胶、龟甲、鹿角与盐酸氨基葡萄糖效果相当,人参、枸杞弱于盐酸氨基葡萄糖。结论:龟甲和鹿角是龟鹿二仙胶抑制凋亡的主要物质。龟甲、鹿角峻补阴阳以生气血精髓的作用可以一定程度上从抑制软骨细胞凋亡来体现。人参大补元气、枸杞滋补肾阴,抑制凋亡有效,但效果明显不如龟鹿二仙胶;而4味药共同作用,具有填补精髓、益气壮阳之功,人参、枸杞可以在一定程度上加强龟甲、鹿角抑制软骨细胞凋亡基因的表达。Objective： To observe the effect of Guilu Erxian decoction and its decomposed recipes on the expression of chondrocyte apoptosis gene of SD rat,and analyze the role of separated compound of the prescription.Methods：The articular cartilage of one month old SD rat was taken to establish chondrocytes culture system in vitro.Real-time quantitative PCR was used to detect mRNA expression of Bax,Bcl-2,Caspase-3,and p53.Results： The mRNA expression of Bax,Bcl-2,Caspase-3,and p53 were inhibited obviously by Guilu Erxian decoction,tortoiseshell,antlers,ginseng,medlar,and glucosamine hydrochloride.The effect of inhibition is equal in Guilu Erxian decoction,tortoiseshell,antlers,and glucosamine hydrochloride,but the effect of ginseng and medlar is lower than that of glucosamine hydrochloride.Conclusion：The tortoiseshell and antlers of Guilu Erxian decoction plays a major role in inhibiting of apoptosis.The role of tortoiseshell and antlers for tonifying Yin and Yang,then leading to induce Qi and blood,can be reflected by its ability of inhibiting chondrocyte apoptosis.The Ginseng which can nourish vitality and medlar which can nourish Yin and tonifying kidney are effective for inhibiting apoptosis,which is obviously lower than Guilu Erxian decoction.The interaction of four herbs can fill with essence,tonifying Qi and nourish kidney Yang.The ginseng and medlar can enhance the effect of tortoiseshell and antlers to inhibit chondrocyte apoptosis.
