摘 要:目的:探讨卷柏水煎液对心肌缺血再灌注损伤(MI/R)的影响及作用机制。方法:KM雄性小鼠随机分为正常对照组、MI/R损伤模型组、卷柏水煎液低剂量+MI/R组、卷柏水煎液高剂量+MI/R组,卷柏水煎液灌胃给药5d后,腹腔注射垂体后叶素和硝酸甘油复制心肌缺血再灌注模型,同时记录不同时点的肢体Ⅱ导联心电图,连续观察80min后,取小鼠心肌组织制作病理切片以观察心肌损伤情况;并测定心肌组织超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量。结果:与正常组相比,模型组小鼠心电图J点发生明显偏移;镜下观察心肌组织病理损伤程度明显加重;SOD活性较正常组降低(23.52±7.21比33.02±4.13,P<0.05);MDA含量较正常组增加(9.80±3.07比5.86±1.04,P<0.05)。与模型组相比,卷柏水煎液预处理组心肌组织病理损伤程度减轻;高剂量组心肌组织SOD活性明显高于模型组(31.96±5.82比23.52±7.21,P<0.05),MDA含量明显降低(5.50±1.80比9.80±3.07,P<0.05);低剂量组SOD活性及MDA含量与模型组比较无显著差异。结论:卷柏水煎液能减轻心肌缺血再灌注损伤,其机制可能与其增加缺血再灌注心肌组织中SOD活性,降低MDA含量有关。Objective:To investigate the effect of selaginellae on myocardial ischemic-reperfusion injury in mice and its mechanism.Methods:Forty male Kunming mice were randomly divided into four groups:normal group,myocardial ischemic-reperfusion injury(MI/R) model group,low dose selaginellae(0.02 ml/g apozem,once a day for 5 days) + MI/R group,and high dose selaginellae(0.02 ml/g apozem,twice a day for 5 days) + MI/R group.MI/R model of mice was established by intraperitoneal injection of pituitrin(20 U/kg) and followed by intraperitoneal injection of nitroglycerin(20 mg/kg) after 30 minutes.Electrocardiogram(ECG,lead Ⅱ) was monitored continuously for 80 minutes.The pathological damage of myocardial tissue was observed under optical microscope after HE staining.Myocardial SOD activity and MDA content were measured by spectrophotometer.Results:Compared with normal control group,J point of ECG raised obviously,the degree of myocardial histopathological injury significantly increased,SOD activity decreased,and MDA content increased in MI/R model group.Compared with MI/R model group,both low and high dose selaginellae pretreatment lowered J point of ECG,and decreased the degree of myocardial histopathological injury.The activity of myocardial SOD increased(31.96±5.82,vs 23.52±7.21 in model group,P0.05),and content of MDA decreased(5.50±1.80,vs 9.80±3.07 in model group,P0.05) in high dose selaginellae group,but low dose selaginellae group had no significant changes in activity of SOD and MDA content.Conclusion:Selaginellae can protect myocardium from ischemic-reperfusion injury maybe through it effects of increasing SOD activity and decreasing MDA content.
