外源性硫化氢对异丙肾上腺素诱导的大鼠急性心肌缺血的保护作用

摘  要:目的观察静脉注射外源性硫化氢(H2S)对异丙肾上腺素(ISO)诱导的大鼠急性心肌缺血的保护作用。方法SD雄性大鼠18只,体重200~250g,随机分为3组(n=6):正常对照组,模型组和NaHS治疗组。模型组和NaHS治疗组采用皮下多点注射ISO方法制作大鼠急性心肌缺血模型。对照组大鼠皮下注射等剂量生理盐水(0.1ml/100g)。NaHS组静脉注射NaHS(50mmol/L,0.1ml/100g),模型组静脉注射等容积生理盐水(0.1ml/100g)。给药后,记录3组大鼠70min内心电图。实验结束后,各组大鼠取血测血清丙氨酸转氨酶(ALT)、天冬氨酸转氨酸(AST)、磷酸肌酸激酶(CK)、磷酸肌酸激酶同工酶(CK—MB)。结果模型组大鼠记录10min后,大鼠心电图sT段明显下降(模型组:-0.21±0.20mV比对照组:0.04±0.04mV,P〈0.05)。静脉注射NaHS能明显改善ISO诱导的大鼠心肌缺血,大鼠心电图ST段在给药后30min开始恢复(模型组:-0.18±0.16mV比NaHS治疗组:-0.09±0.06mV,P〉0.05),50min内基本恢复正常(NariS治疗组:-0.01±0.06mV比对照组:0.01±0.05mV,P〉0.05),和模型组比较有显著性差异(模型组:±0.19±0.17mV比NaHS治疗组:-0.01±0.06mV,P〈0.05)。NaHS治疗组血清ALT、AST、CK、CK—MB与模型组比较显著降低。结论外源性硫化氢对异丙肾上腺素诱导的大鼠急性心肌缺血有显著的保护作用。Objective To observe the protective effect of hydrogen sulfide on the acute myocardial ischemia induced by isoprenaline (ISO) in rats. Methods Eighteen male SD rats, weighing 200 - 250g, were randomized into 3 groups ( n = 6) : control group, model group and NariS treatment group. Acute myocardial ischemia was induced by multi - point subcutaneously injection of ISO in rats of model and NariS group. Saline (0. lml/100g) or NariS (50mmol/L, 0. 1ml/100g) was administrated comparatively in control group or NariS group. Electrocardiogram (ECG) was recorded in 70rain. Serum alanine aminotrasferase (ALT) , aspartate aminotrasferase (AST) , creatine kinase(CK) and creatine kinase isoenzyme( CK - MB) were determined. Results After injection of ISO 10mins, ST segment of rats dereased obviously in model group than that in control group( P 〈 0.05 ). ST segment began to rise after 30min and recovered in 50min(P 〈 0. 05 , compared with model group). The serum levels ALT, AST, CK and CK - MB were lower in Naris treatment group than those in model group. Conclusion Hydrogen sulfide effectively ameliorates acute myocardial ischemia induced by ISO of rats.
