摘 要:目的:建立高效液相色谱测定南方红豆杉种子中紫杉醇含量的方法。方法:采用WhatmanC18(250mm×4.6mm,5.0μm)色谱柱,柱温30℃;流动相为乙腈-水(35:65-80:20)30min梯度洗脱,流速1.0mL/min;检测波长227nm。结果:紫杉醇的线性范围:8.5~51.0μg/mL(r=0.9996),平均回收率为99.7%,RSD为1.57%;三批南方红豆杉种子种紫杉醇的含量分别为:0.0196%、0.0212%和0.0205%。结论:方法稳定可靠,重复性和精密度良好,可作为南方红豆杉药材的含量测定方法,为紫杉醇的分离纯化及该药材的质量控制提供科学依据。Objective: To establish a method to determinate the contents of Taxol in Taxus mairei seeds by HPLC. Methods: Whatman C18(250 mm×4.6 mm, 5.0 μm)was used, the mobile phase was composed ofacetonitrile-water (35:65 - 80:20); The flow rate was 1.0 mL/min; Column temperature was 30℃ ;Taxol's detection wavelength was 227 nm. Results: The linear range of Taxol was at 8.5 - 51.0 μg/mL (r = 0.999 6). The average recovery was 99.7% ,RSD = 1.57% ;The three contents of taxol in Taxus mairei seeds were 0.019 6%, 0.021 2% and 0.020 5% respectively. Conclusion: The results proved that the method was reliable with good repeatability and precision and can be used for the content determination of the crude drugs of Taxus mairei. The results also provided the scientific evidence for purification and quality control of Taxol.
