摘 要:采用基因测序的方法,对6种鳗鲡的COⅡ基因片段进行了PCR扩增和测序。结果表明,6种鳗鲡COⅡ基因片段的长度都为412bp,4种碱基组成非常相似,并且A+T的含量都大于G+C的含量。6种鳗鲡COⅡ基因片段核苷酸序列之间有46bp的差异,其中有6处发生碱基颠换,40处发生碱基转换。非洲鳗鲡(Anguilla mossambic)和日本鳗鲡(A.japonica)COⅡ基因片段核苷酸序列差异最大为27bp,同源性为93.45%,而日本鳗鲡和菲律宾鳗鲡(A.mormorata)COⅡ基因片段核苷酸序列差异最小为5bp,同源性为98.79%。MtDNA COⅡgene fragments of six Anguilla species were amplified using the PCR method, and the products were purified and sequenced. The results showed that all the mtDNA COⅡgene fragments had 412bp with similar base composition, which had greater amount of the A+T pairs than the G+C pairs. The difference among the six mtDNA COⅡgenes was 46bp, including 6 base transversions and 40 base transitions. The difference between the mtDNA COⅡ genes of A. mossambica and A. japonica was 27bp with 93.45% homology; however the difference between those of A. japonica and A. mormorata was 5bp with 98.79% homology.
