摘 要:目的:观察丹参川芎嗪合剂腹腔注射给药对博来霉素(bleomycin,BLM)致肺纤维化大鼠肺组织Ⅰ、Ⅲ型胶原mR-NA表达的影响。方法:健康成年SD雄性大鼠90只,随机分为生理盐水(NS)组,BLM组,地塞米松(D)组,丹参川芎嗪合剂小剂量(S)组,合剂中剂量(M)组,合剂大剂量(L)组,每组15只,后五组按照BLM 5mg/kg气管内给药制备肺纤维化大鼠模型,NS组气管内灌注同体积的NS。24小时后S组、M组、L组分别腹腔注射丹参川芎嗪合剂药物,NS组、BLM组腹腔注射等体积的NS,连续给药至第7天,第14天,第28天后各组分别处死大鼠5只。取肺组织分别行HE和Masson染色,观察肺泡炎和肺纤维化情况并评分;实时荧光定量PCR(Real-time quantitative Polymerase Chain Reaction,Real Time PCR)测定肺组织Ⅰ、Ⅲ型胶原mRNA的表达;血清用于肝、肾功能检测。结果:(1)各组大鼠血清肝、肾功能测定结果两两比较差异无统计学意义。(2)肺组织病理学观察提示:NS组各时间点大鼠未发现肺泡间隔水肿、炎性细胞浸润及肺组织纤维化的形成;BLM组第7天大鼠肺泡炎症改变明显,肺组织纤维化不明显,BLM组第14天、第28天大鼠肺泡结构破坏、间质增生,形成广泛纤维化,同时存在肺泡炎;D组有肺泡炎症改变,但是炎性细胞浸润程度较BLM组轻,纤维化程度和肺泡结构破坏也较BLM组轻;S组肺泡炎和肺纤维化程度均与BLM组类似。M组、L组第7天大鼠肺组织改变与BLM组大致相同;M组、L组第14天肺泡炎程度明显低于BLM组。M组、L组第14天、第28天肺纤维化程度明显低于BLM组(P<0.05)。(3)实时荧光定量PCR检测肺组织Ⅰ、Ⅲ型胶原mRNA表达:以NS组为参考标准,BLM组mRNA表达明显增多,D组相对于BLM组表达明显减少;S组与BLM组比较无明显差异,与D组比较其表达增多;M组、L组明显低于BLM组,与S组比较表达也明显减少(均P<0.05)。结论:丹参川芎嗪合剂腹腔注射能够改善肺纤维化,且对�To observe the effects of Salvia and TMP mixture on the mRNA expression of type I and In collagen in lung tissue of bleomycin (BLM) induced pulmonary fibrosis rats. Methods: 90 adult male SD rats were randomly divided into six groups: normal saline (NS) group, BLM group, dexamethasone (D) group, small dose of Salvia TMP mixture (S) group, medium dose of Salvia TMP mixture (M) group, and large dose of Salvia TMP mixture (L) group. Intratracheal instillation of BLM was given to BLM group, D group, S group, M group and L group, and NS was given to NS group. After 24 hours, S group, M group and L group were intraperitoneally in- jected Salvia TMP mixture, NS group and BLM group were intraperitoneally injected normal saline. Five rats in each group were killed on day 7, 14, 28. HE and Masson staining were performed to observe alveolitis and pulmonary fibrosis. Real-time quantitative PCR technology was used to detect mRNA expression of type I and m collagen. Serum was used to detect liver and kidney function. Results: (1) The results of liver and kidney function in each group had no significant difference. (2) Alveolar septal edema, inflammatory cell infiltration and fibrosis were not found in NS group; alveolitis existed in D group, but infiltration of inflammatory cells was less than BLM group, and collagen fibers and alveolar struc- tural damage also less; alveolitis was significant and fibrosis was not obviously observed in BLM group on day 7. On the contrary, fibrosis was significant and alveolitis was not obviously observed in BLM group on day 14 and 28; the degree of alveolitis and pulmonary fibrosis in S group were similar with BLM group; the diversification in lung tissue in M group and L group on day 7 was roughly the same as BLM group, and the degree of alveolitis in M group and L group on day 14 was significantly lower than that of BLM group; the degree of pulmonary fibrosis in M group and L group on day 14 and 28 was significantly lower than that in BLM group(P〈0.0
