化瘀泄浊方对肾间质纤维化实验大鼠整合素连接激酶的影响

摘　　要：目的:观察化瘀泄浊方对肾间质纤维化实验大鼠整合素连接激酶(ILK)的影响。方法:选用健康、雌性SD大鼠50只,用单侧输尿管结扎方法复制大鼠肾间质纤维化模型,将其随机分为假手术组、化瘀泄浊方组[化瘀泄浊方5.7g/(kg.d)经口灌服]、阿魏酸哌嗪分散片组[给予阿魏酸哌嗪分散片6.7mg/(kg.d)]、缬沙坦组[给予缬沙坦5.3mg/(kg.d)经口灌服]及模型组[给予同等量生理盐水经口灌服],每组10只大鼠。全部大鼠于实验第21天全部杀检,免疫组化及RT--PCR检测肾组织ILK蛋白及mRNA的表达。结果:免疫组化及RT-PCR结果显示,与假手术组比较,模型组和各治疗组ILK的表达显著增强,差异有统计学意义(P<0.05);与模型组比较,化瘀泄浊方组、阿魏酸哌嗪分散片组、缬沙坦组ILK的表达减弱,差异有统计学意义(P<0.05),其中化瘀泄浊方组ILK的表达优于阿魏酸哌嗪分散片组,差异有统计学意义(P<0.05),其他各组间均无明显差异(P>0.05)。结论:化瘀泄浊方能通过抑制肾组织ILK蛋白及其mRNA的表达,使病变肾组织细胞外基质合成减少,发挥对肾间质纤维化的治疗作用。Objective： To observe the effect of Hua-Yu-Xie-Zhuo-Recipe on integrin linked kinase（ILK） in rats with tubulointerstitial fibrosis.Methods： The model of renal obstruction was established by unilateral ureteral obstruction（UUO） in rats and then divided into five groups,including sham-operation group,UUOZY group[treated with treated with Chinese medicine of huayuxiezhuo 5.7g/（kg·d）],UUOA group[treated with ferulaic acid piperazidine dispersible tablet 6.7 mg/（kg·d）],UUOV group[treated with Valsartan 5.3mg/（kg·d）] and.UUO group,The protein and mRNA of ILK in kidney tissue were observed.Results： The expression of ILK of UUO and the treatment groups was increased significantly compared with sham group（P〈0.05）;the expression of ILK of all treated groups declined notablely compared with model group（P〈0.05）.Hua-Yu-Xie-Zhuo-Recipe group was superior to piperazine ferulate dispersible tablets group（P〈0.05）.There was no markedly difference among other groups（P〉0.05）.Conclusion： Hua-Yu-Xie-Zhuo-Recipe can inhibit the expression of ILK and ILK-mRNA in renal tissue,resulting reduction of ECM.So,Hua-Yu-Xie-Zhuo-Recipe embody its therapeutic effect on renal interstitial fibrosis.
