一种简便、快速从福尔马林固定石蜡包埋组织块提取HIV前病毒DNA的方法

摘　　要：把5μm厚福尔马林固定石蜡包埋的AIDS患者病理组织切片以蛋白酶K消化后加热,经乙醇沉淀后所产生的DNA直接用于PCR扩增HIV前病毒DNA,其产物经溴化乙锭染色后全部可以看到阳性条带。此技术快速、简单、实用,虽经减化抽提步骤,但并不影响对HIV片段的扩增。DNA was extracted from 5μm sections of neutral buffered 10% formalinfixed, paraffin embedded tissue by proteinase K digestion, following by boiling. After ethanol precipitation, the extracted DNA was subject to polymerase chain reaction (PCR) for amplification of provirus of HIV. Ethidium bromide stained PCR product could be visualized in agarose gel. This is a simple, rapid and practical technology, although the DNA extraction procedure is simplified, the amplification to DNA fragments won't be influenced.
