摘 要:目的采用响应面法优化关白附多糖的提取工艺。方法根据单因素试验结果,选择提取温度、液料比和提取时间作为响应因子,多糖得率为响应值,进行三因素三水平的响应面试验,通过建立的二次多项式数学模型,研究各因素对响应值的影响,得到关白附多糖的最佳提取工艺。结果优化后关白附多糖的最佳提取工艺为:提取温度81.37℃,料水比32.70∶1 mL/g,提取时间3.43 h,此时关白附多糖的理论预测值为26.39%,最优条件下多糖得率的实验值为26.82%,与理论预测值的相对误差为1.65%。结论采用响应面分析法优化关白附多糖的水提工艺,具有实际应用价值。Objective To optimize the extraction conditions of polysaccharide in Aconitum coreanum using response surface methodology(RSM).Methods Extracting temperature,solvent/solid ratio and extracting time were selected as response factors.The yield of polysaccharide was used as response value.Base on the single factor experiments,a three-factor-three-level experiment design was developed by Box-Benhken central composite design method.The response surface method was employed to analyze the results of experiments.Results The optimum extraction conditions of polysaccharide were as follow: extracting temperature was 81.37 ℃,solvent/solid ratio was 32.70:1(mL/g) and extracting time was 3.43 h.The predictive maximum yield of polysaccharide was 26.39 %.The average yield of polysaccharide in 3 validation experiments was 26.82 %.The relative error was 1.65 %.Conclusion RSM analysis employed in optimization of the extraction conditions for polysaccharide in Aconitum coreanums is feasible.
