石见穿萃取物对体外培养成骨细胞活性的影响  

Effects of Extract from Salivia chinensis Benth on Proliferation and Activity of Osteoblastic MC3T3-E1 Cell in Vitro

摘  要:石见穿以95%乙醇抽提得总提物.分别用不同试剂萃取总提物,获得其石油醚萃取物、乙酸乙酯萃取物、正丁醇萃取物以及其水溶性成分.以体外培养成骨细胞MC3T3-E1为模型,用cck-8法测定细胞增殖,碱性磷酸酶(ALP)试剂盒法测定了上述不同萃取物对成骨细胞增殖和分化的影响.结果表明:总提物在浓度10-3、10-4、10-5mg/mL时和10-3mg/mL乙酸乙酯萃取物具有极显著的促进细胞增殖(P<0.01)作用.10-4mg/mL总提物和10-3mg/mL乙酸乙酯萃取物显著促进成骨细胞ALP活性(P<0.05).研究表明石见穿乙酸乙酯萃取物含有能促进成骨细胞增殖和成骨活性的有效成分.Salivia chinensis Benth were extracted with different solvents and the effects of the extracts on the proliferation and differentiation of osteoblast were investigated.Salivia chinensis Benth was first extracted with 95 % ethanol followed seperately by extracted with petroleum ether,ethylacetete or butyl alcohol.MC3T3-E1 osteoblast-like cells were used as in vitro model to test the effects of different fractions on osteoblast proliferation and differentiation.It was found that the general extract of 10-3,10-4,10-5 mg/mL and the ethylacetete fraction of 10-3 mg/mL could stimulate cell proliferation(P0.01).Moreover,the general fraction of 10-4 mg/mL and ethylacetete fraction 10-3 mg/mL enhance alkaline phosphatase(ALP) activity of MC3T3-E1(P0.05).It is conclusion that the components in the ethylacetate fraction from Salivia chinensis Benth can stimulate osteoblast proliferation and differentiation.
