摘 要:通过MTT(四甲基偶氮唑盐)法绘制细胞增殖曲线观察药物对肝癌细胞系HepG-2和食管鳞癌细胞系EC-1细胞增殖的影响,用荧光染色法进一步确定"龙泉复方"有效成分诱导细胞凋亡的较佳配伍及其有效作用浓度范围.结果表明:对抑制HepG-2细胞增殖有效作用的浓度:七味单方(马钱子、山慈菇、喜树果、龙葵、石上柏、青黛、紫菀)浸膏混合物和龙葵单方浸膏均〈125μg/mL,七味药物复方总浸膏为250~1000μg/mL;对抑制EC-1细胞增殖有效作用的浓度:三味药物混合浸膏(马钱子,喜树果、青黛提取的浸膏的混合物)〈50μg/mL和150~400μg/mL,七味单方混合浸膏〈25μg/mL和50~400μg/mL,喜树果单方浸膏为25μg/mL和50~200μg/mL;3种浸膏促进EC-1细胞增殖的浓度依次为:50~100、25~50、25~50μg/mL.通过对EC-1荧光染色观察发现诱导细胞凋亡的有效作用浓度:七位单方浸膏混合物为5~10μg/mL,三味药复方混合浸膏为10~25μg/mL,喜树果约为50μg/mL.研究结果提示,药物浓度范围和细胞系的不同导致其效果也不尽相同,临床应据具体情况调整,使之用药少效果佳.The cell proliferation curve and observation on the effect of the cells of HepG-2 and EC-1 caused by the drugs were obtained through the method of MTT(methyl thiazolyl tetrazolium),and the optimal compatibility and concentration of the drugs were determined by fluorescence staining.It is shown that the seven-mixed drug(the major effective components extracted from strychnine,mountain keats mushroom,common camptotheca fruit,solanum nigrum,stone cypress,natural indigo and tatarian aster root) and solanum nigrum are both effective below 125 μg/mL,and the range for the seven-drug mixture(mixed before extraction) is between 250-1000 μg/mL for HepG-2 cell.While for EC-1,the effective ranges of the three-mixed drugn(the major effective components extracted from strychnine,common camptotheca fruit and natural indigo) are below 50 ?g/mL and between 150-400 μg/mL;the seven-mixed drug's ranges are below 25 μg/mL and between 50-400 μg/mL;for common camptotheca fruit,the effective ranges are below 25 μg/mL and between 50-200 μg/mL.However,the concentrations of those three different drugs which caused prolification of EC-1 are between 50-100 μg/mL,25-50 μg/mL and 25-50 μg/mL,respectively.From the results of the more accurate method of fluorescence staining,it is found that the ranges for the seven-mixed drug;the three-mixed drug;and the common camptotheca fruit are 5-10 μg/mL,10-25 μg/m and about 50 μg/mL respectively.The selected compatibility showed obvious inhibition on cancer cells.The results indicate that different concentrations and cancer cells lead to different effect,the drugs and concentrations should be chosen and adjusted in different condition in order to obtain good effect with less amounts.
