摘 要:通过Y3、Y4与优良亲本配制杂交组合,探索转Bt基因甘蔗杂交利用的可能性。采用PCR和Western Blot方法对杂交后代进行分析,结果表明,被测定的12个杂交组合的641株实生苗中,有45株含Bt基因,占7.02%,转Bt基因甘蔗育性及其杂交亲和力偏低。PCR检测结果与Western Blot分析结果高度一致。田间试验表明,绝大多数低代Bt基因甘蔗品系在农艺、经济性状方面,与优良栽培品种相比仍存在较大差距。建议加强优良Bt甘蔗品系的杂交利用。Two Bt-transgenic sugarcane lines, Y3 and Y4, were crossed with the local elite parents, in order to investigate the possibility of utilization of Bt-transgenic sugarcane lines in sugarcane Breeding. PCR and Western Blot technology were employed to analyze the progenies from these crosses. In total, 12 cross combinations were done and produced 641 seedlings, of which 7.02%, i.e. 45 seedlings, were Bt-transgenic plants. In the present research, the result of distinguishing Bt-transgenic plants from other progenies using PCR method was consistent with that using western Blot analysis. Since PCR method is easier and simpler comparing with western blot analysis, PCR was an efficient method for inspecting Bt-transgenic sugarcane. On the other hand, our research results also showed that the low fertility and crossing-compatibility existed in Bt-transgenic sugarcane. The result of the field trial also showed that most of Bt-transgenic sugarcane progenies' agronomic and economic traits were worse than the elite varieties. To sum up, the utilization of Bt-transgenic sugarcane lines should be strengthened in the future in sugarcane breeding.
