摘 要:对烟台套子湾海域2009年5~8月份爆发增殖的大型绿藻进行了监测,对优势种ITS区进行扩增测序和分析鉴定。结果表明:5月18日~6月10日,套子湾绿藻优势种为石莼类2009Ulva-A;6月10日~7月5日,绿藻优势种为浒苔类2009Ente—B;7月5日~8月初,绿藻优势种为刚毛藻2009Chla—C。三株绿藻分别扩增得到491bp,483bp和516bp长ITS序列,与其他18种绿藻相应ITS序列比对,计算遗传距离,构建系统发生树,2009Ulva—A、2009Elate—B和2009Chla-C判定分别为长石莼(Ulvalinza)、肠浒苔(Enteromorpha intestinalis)和刚毛藻(Cladophorasp)。弄清套子湾大型绿藻优势种分类地位,研究绿藻生消规律,为了解套子湾大型绿藻过量增殖原因及绿潮防治提供理论依据。The species of Chloraphyta were monitored from May to Aug,2009 when the algae may outbreak large-scale in Yantai Cop Bay, and ITS sequences of the dominant species were amplified, sequenced and identified. The dominant species was Ulva from May 18 to Jun 10 and named 2009 Ulva-A. The dominant species changed to Enteromorpha, 2009 Ente B, from Jun 10 to Jul 5. After that, Cladophora (2009 Chla-C) became the dominant species from Jul 5 to Aug 30. 491 bp,483 bp and 516 bp of ITS sequences were amplified from 2009 UIva-A, 2009 Ente-B and 2009 Chla-C. By blastn analysis, phylogenetic tree construction and genetic distance calculation with other 18 Chloraphyta sequences, 2009 Ulva-A, 2009 Ente B and 2009 Chla-C were identified as Ulva linza, Enteromorpha intestinalls, and Cladophora sp. The clearing taxonomic status and studying the variation of dominant species of Chloraphyta in Cop Bay may help to make out the reason of green tide and provide the theoretical basis for prevention and treatment.
