摘 要:研究血管紧张素原(AGT)基因M235T多态性对NIDDM患者发生高血压的影响。方法用限制性酶谱分析法检查AGT基因M235T多态性M235和T235的频率和TT、TM和MM基因型在正常对照组(68例)、糖尿病非高血压组(104例)和糖尿病高血压组(87例)中的分布,并检查各基因型对实验对象血压水平的影响。结果糖尿病高血压组患者AGT基因M235T多态性T235的频率明显高于对照组和糖尿病非高血压组。TT基因型在糖尿病高血压组中的比例明显高于对照组和糖尿病非高血压组。TT基因型者的收困压和舒张压显著高于TM和MM基因型者。结论携带AGT多态性TT基因型者的收缩压和舒张压水平显著高于携带TM和TT基因型者;血管紧张素原基因M235T多态性的T235变异是NIDDM合并高血压的危险因素。Objective To investigate if M235T polymorphism of angiotensinogen gene has any effect on the occurrence of hypertension in Chinese NIDDM patients. Methods By the method of restriction enzyme mapping, we investigated the allele frequencies of T235 and M235 and the proportions of the three genotype MM, TM and TT of the polymorphism of angiotensinogen gene in the group of normal controls (n=68).diabetic normotension (n = 104) and diabetic hypertension (n = 87). Results The group of diabetic hypertension had a significant higher al1ele frequency of T235 (P<0. 001 ) and a significant higher (P<0.01) proportion of TT genotype of the polymorphism of angiotensinogen gene. The subjects who had the genotype of TT had significant (P<0.05) higher systolic blood pressure and diastolic blood pressure levels than those who had the genotype of MM and TM. Conclusion These data provided the evidence in favor of an association between hypertension in Chinese NIDDM patients and the variant of T235 of the polymorphism M235T of angiotensinogen gene.
