摘 要:目的探讨干扰素(Interferon,IFN)对ICR乳鼠感染肠道病毒71型(Enterovirus 71,EV71)的保护作用。方法将FY23-EV71感染经重组人IFNα1b和IFNλ1预处理的Vero和KMB17细胞,微量滴定法检测病毒滴度;取ICR乳鼠分为PBS对照组、EV71对照组和IFN保护组,临床观察2周,取乳鼠组织,观察病理学变化,并检测病毒含量。结果两种IFN对KMB17细胞和Vero细胞感染EV71均有保护作用,且IFNα1b的保护作用优于IFNλ1;IFN保护组乳鼠较EV71对照组乳鼠在临床表现、体重、病理变化和组织病毒含量等方面均表现出对EV71感染的保护作用。结论 IFN能抑制EV71的增殖,并对病毒致病性和组织嗜性有一定的影响。Objective To investigate the protective effect of interferon(IFN) against enterovirus 71(EV71) infection in suckling ICR mice.Methods Vero and KMB17 cells previously treated with recombinant human IFNα1b and IFNλ1 were infected with FY23-EV71 and determined for virus titer by microtitration.Suckling ICR mice were infected with EV71 and treated with IFN,using those infected with PBS as negative control,and those untreated with IFN after EV71 infection as positive control,and observed for clinical signs for 2 weeks.The brain tissues of mice were collected,observed for pathological change,and determined for virus load.Results Both IFNα1b and IFNλ1 showed protective effect against EV71 infection in KMB17 and Vero cells,while the protective effect of IFNα1b was superior to those of IFNλ1.IFNα1b showed protective effect against EV71 infection in suckling mice,as proved by clinical sign,bodyweight,as well as pathological change of virus load in brain tissue.Conclusion IFN inhibited the replication of EV71,and showed a certain effect on pathogenicity and tissue tropism of EV71.
