摘 要:目的:探讨制备小鼠膀胱整标本的方法.以及整标本技术在尿路上皮形态学中的应用。方法:取C57BL/6小鼠膀胱组织制作整标本.对制作完成的标本进行切片核染色和整标本核染色。用解剖显微镜.表面荧光显微镜和共聚焦荧光显微镜观察整标本膀胱的大体和超微形态.并建立小鼠膀胱上皮的3D成像。结果:小鼠膀胱整标本组织形态良好.膀胱上皮完整.表面平坦.黏膜皱襞全部消失。共聚焦荧光显微镜逐层扫描可观察到膀胱上皮全层及上皮下间质层。结论:小鼠膀胱整标本的制备简便易行.可弥补连续切片无法达到的组织整体观。可以用于组织形态学的分析研究.对于膀胱上皮发生学、生理学和病理生理学的研究具有十分重要的意义。Objective:To establish, the whole mount technique of mouse bladder, and to explore its application in urothelial morphology. Methods:C57BL/6 mouse bladders were used to produce whole mount samples. Frozen sections were produces as well Both the whole mount bladders and sections were then processed to nuclear staining. Stereo microscopy, epifluorescence microscopy, and confocal microscopy were used to study the gross morphology and ultrastructure of bladder whole mounts, and the data was reestablished by 3D reconstitution. Results: The morphology of mouse bladder whole mounts was well preserved, and bladder epithelium was intact with smooth surface. All of the ridges in bladder lumen disappeared. The whole bladder epithelium and part of the mesencbymal layer could be reached by z-axis scanning using confocal microscopy. Conclusions: Manual method for the preparation of mouse bladder whole mounts is an easy and effective technique. It can provide three-dimensional in- formation, which is unavailable merely by serial sections. It could be used in histological and morphological studies, which is highly significant in the phylogenetics, physiology, and pathophysiology of bladder epithelium.
