Nodosin预处理移植肝对移植早期原位细胞因子格局的影响

摘  要:为研究溪黄草有效成分Nodosin预处理供肝后,细胞因子分泌格局在大鼠移植肝中的变化。将24例Wister→SD大鼠原位肝移植模型随机分成四组,分别为未处理对照组、Nodosin组、Copp组和Znpp组,分别在移植手术后第1和第4天切取肝脏组织,采用病理学和细胞生物学方法比较不同组别肝脏病变,定量PCR方法检测肝脏内细胞因子的表达。结果显示,供肝保留灌注Nodosin后,可以降低供肝组织内的细胞凋亡率、肝脏病理性损伤和坏死;移植原位肝脏中TNF-α和IFN-γ的表达在Nodosin处理中的表达明显下降(P<0.05);IL-10表达增高,由肝细胞分泌的内皮素的含量也有下降。结果表明,Nodosin灌注预处理可以在移植早期明显降低Th1型细胞免疫应答,同时降低肝组织内皮素含量,提示Nodosin可能通过改变肝移植物周围免疫微环境,促进移植肝脏的存活和功能,从而提供原位肝移植的有效性。To study the cytokine profiles in orthotopic transplanted liver pretreated with reserved infusion of Nodosin in rat,24 male Wistar-SD rats were randomly assigned into 4 groups(n=3).i.e.control group,Nodosin group,positive control group(Copp group) and negative control group(Znpp group).Hepatic tissues were obtained 1 day and 4 days after perfusion.Histochemical,biochemical and Tunel assays were performed to evaluate the efficacy of liver transplantation.Four correlation factors(TNF-α,IFN-γ,IL-10 and ET-1) were detected by RT-quantitative PCR.After the donor livers were reservedly infused with Nodosin,liver cell apoptosis was less than that in control group.Serum level of AST was also dramatically decreased(P0.05) compared to the control group as well as Suzuki's score in representative rat liver recipients.More importantly,cytokines involved in the acute rejection including TNF-α and IFN-γ were less expressed in Nodosin-treated group than in control group(P0.05) whereas the level of IL-10 with immuregulatory role in the liver was increased.Endothelin-1 was down-regulated in Nodosin-pretreated livers after transplantation.Our results demonstrated that reserved infusion with Nodosin before liver transplantation could effectively attenuate the apoptosis and liver injury of the implanted liver.This was consistent with the shifts of cytokine expression from Th1 rejection responses to regulatory immune responses.It is evident that these effects together with the amelioration of liver function mediated by Nodosin would improve the survival and function of implanted liver,resulting in the efficacy of liver transplantation.
