摘  要:为了观察Toll样受体1(TLR1)及FasL在大鼠哮喘中的作用及布地奈德(BUD)对其的影响,采用大鼠哮喘模型,随机分成正常对照组、哮喘组、布地奈德治疗组为研究对象,取外周血,流式细胞仪检测其淋巴细胞上TLR1和FasL的表达水平,右肺行病理学检查。结果发现:哮喘组外周血淋巴细胞TLR1、FasL表达水平均显著高于对照组(P<0.01),且与BUD组比较TLR1、FasL表达有显著性差异(P<0.01)。流式细胞散点图提示哮喘组淋巴细胞TLR1呈高表达。在哮喘组和BUD组以及正常对照组外周血淋巴细胞FasL表达均较低,但是,在哮喘组仍明显高于正常对照组(P<0.01)。提示:淋巴细胞上TLR1和FasL的表达水平在哮喘时增加,TLR1、FasL参与了哮喘的发病机制,TLR1在哮喘的气道慢性炎症的发生和发展中起重要作用,而哮喘组存在淋巴细胞凋亡下降,可能是哮喘气道慢性炎症持续存在的原因。作者认为BUD吸入有可能通过调节淋巴细胞凋亡的平衡来治疗哮喘。To observe the expressions of toll-like receptor 1(TLR1) and Fas and their influence by Budesonide(BUD),the rat model of asthma were randomly divided into three groups.i.e.asthma group,control group and BUD-treated group.Flow cytometry was used to analyze the expression of TLR1 and FasL on lymphocyts of peripheral blood.Rats were sacrificed and their right lung tissues were undergone for pathological examination.It was demonstrated that the expression levels of TLR1 and FasL in asthma group were significantly increased than that in control group(P0.01).In addition.they were significantly decreased in BUD group than in asthma group(P0.01).The asthma group showed a high expression of TLR1 by flow cytometry plot,and FasL expression in peripheral blood lymphocytes were lower in all groups.However,in the asthma group,the level of FasL was significantly increased than that in control group(P0.01).Levels of TLR1 and FasL were increased in rat model of asthma.From these observations,it appears that TLR1 and FasL may be involved in the pathogenesis of asthma.in which TLR1 plays an important role in chronic airway inflammation and development of asthma.Meanwhile the apoptosis of lymphocytes from asthma group was found to be decreased and the inhibited apoptosis might be one of reasons for the chronic persistence of inflammation responses in asthma.BUD inhalation can be used as a measure to treat asthma through regulating the balance of lymphocyte apoptosis.
