摘  要:目的探讨铁调素对骨骼肌铁代谢的调节机制。方法培养大鼠骨骼肌L6细胞,随机分成4组,每组6个重复,每组各加入不同浓度的铁调素溶液(终浓度分别为0、0.05、0.1、0.15 mg/L)孵育12h,采用同位素示踪法,用55Fe测定细胞铁摄取及铁释放能力的变化;通过流式细胞仪检测细胞内铁池(LIP)铁含量;应用免疫印迹法检测膜铁转运蛋白1(FPN1)的变化。结果与空白对照组比较,0.1mg/L铁调素处理组肌细胞的铁释放能力明显下降(P<0.05),0.05及0.15mg/L铁调素处理组肌细胞的铁摄取和铁释放均无明显变化(P>0.05);0.1mg/L铁调素处理组其细胞内LIP铁含量较对照组明显升高(P<0.05);0.1mg/L铁调素处理组细胞膜上FPN1的表达较对照组明显降低(P<0.05)。结论铁调素可通过影响骨骼肌细胞FPN1的表达水平,降低骨骼肌细胞内铁的向外转运,增加细胞内铁池的铁含量,从而调节骨骼肌细胞的铁代谢。Objective To illuminate effects of hepcidin on iron metabolism of skeletal muscle. Methods L6 rat skeletal muscle ceils were randomly divided into four groups and there were 6 repeats in each group. The cells were incubated with hepcidin at different concentrations (0, 0. 05 rag/L, 0. 1 mg/L, 0. 15 mg/L) for 12 hours. Isotopic tracer method was used to investigate the change of cell iron uptake and iron release. The content of iron in liabile iron pool(LIP) was investigated by flow cytometry and the expression of ferroportin 1 ( FPN1 ) in rats' gastrocnemius was observed by Western blotting. Results Compared with the control group, the released iron of L6 ceils which were incubated with 0. 1 mg/L hepeidin decreased significantly ( P 〈 0.05 ). There was no significant difference of iron uptake and iron release after incubated with 0.05 and 0. 15 mg/L hepcidin ( P 〉 0.05 ). The LIP of L6 cells which were incubated with 0. 1 mg/L hepcidin increased significantly (P 〈 0. 05). The expression of FPN1 decreased significantly ( P 〈 0. 05 ). Conclusion Hepcidin might play an important role in iron metabolism of skeletal muscle cells, which can decrease the expression of FPN1. As a result, the released iron of skeletal muscle cells is reduced and the iron content of iron pool increases.
