摘 要:目的观察人钙离子激活的氯离子通道1(hCLCA1)在慢性鼻窦炎筛窦黏膜中的表达,探讨其在慢性鼻窦炎发病机制中的作用。方法取慢性鼻窦炎患者筛窦黏膜10例,另取10例正常筛窦黏膜作为对照。应用实时定量聚合酶链反应(RT-PCR)方法检测各组鼻窦黏膜中hCLCA1mRNA的表达,应用免疫荧光方法观察hCLCA1蛋白在各组鼻窦黏膜中的表达,比较hCLCA1mRNA及蛋白在各组中的表达情况。结果 hCLCA1表达于鼻窦黏膜上皮的杯状细胞。慢性鼻窦炎组hCLCA1mRNA表达水平及阳性细胞面积比(51.6%±4.5%)高于对照组(7.2%±5.1%),差异有统计学意义(P<0.01)。结论在基因和蛋白水平上,hCLCA1在慢性鼻窦炎鼻窦黏膜中均有明显表达,其在慢性鼻窦炎的发病机制中可能具有重要作用,可能与上皮杯状细胞的增生及黏液高分泌有关。Objective To examine the expression of human calcium-activated chloride channe 1 (hCLCA1) messenger RNA (mRNA) and proteins in human sinus mucosa of chronic sinusitis. Methods Ten ethmoid sinus mucosa was harvested from patients undergoing endoscopic sinus surgery for chronic sinusitis(CRS) and 10 for non-CRS pathologies (control). Then sinus mucosa was analyzed using real-time polymerase chain reaction (RT-PCR) to detect the mRNA of hCLCA1. Immunofluorescent staining was used to evaluate the expression of hCLCA1 proteins in the sinus mucosa. The mRNA and protein expression levels in the epithelia were compared between the CRS and the control group. Results The expression of hCLCA1 mRNA in the sinus mucosa of CRS significantly increased compared with that in the normal sinus mucosa (P 〈 0.01 ). hCLCA1 proteins were expressed in the goblet ceils. Area ratio of positive cells in the epithelia was found higher in the CRS group compared with that in normal group (P 〈 0.01 ). Conclusions This study showed that hCLCA1 gene was up-regnlated in sinus mucosa of CRS. hCLCA1 might play an important role in the pathogenesis of CRS for the over-production of mucus.
