摘  要:目的:观察大鼠脂肪源性干细胞(ADSCs)向成脂诱导分化的生物学特性,以探讨ADSCs在治疗盆腔脏器脱垂(POP)中生物工程学的意义。方法:酶消化法体外分离培养大鼠ADSCs;相差显微镜观察ADSCs的细胞形态;运用流式细胞术鉴定细胞周期以及细胞表面抗原的表达;对ADSCs进行成脂诱导分化,油红"O"染色观察其向脂肪细胞分化的能力。结果:体外培养的ADSCs增殖迅速,呈成纤维细胞样生长,细胞呈多角形、长梭形,形态均一;细胞周期结果显示G0/G1期细胞占90.60%;表面抗原阳性表达率分别为CD44 99.88%,CD90 99.74%,CD105 95.00%,CD49d 89.56%,CD34 3.04%,CD106 0.85%,CD45 0.26%;成脂诱导实验细胞内出现脂滴,诱导14天后油红"O"染色呈橘红色。结论:ADSCs易于分离培养,具有向成脂诱导分化的功能,提示ADSCs可作为治疗POP的种子细胞。Objective:To observe the biological characteristics of the adipose-derived stem cells(ADSCs) differentiated into adipogenic cells,to investigate the feasibility of ADSCs as the seed cells in treatment of pelvic organ prolapse(POP).Methods:ADSCs were isolated and cultured in vitro,and the morphological changes were observed by phase-contrast microscope.The expressions of surface antigenes and the cell cycle were detected by flow cytometry.After induced by adipogenic inducing factor,the differentiations to adipocytes were verified by oil red "O" staining.Results:ADSCs propagated rapidly in vitro,and constituted a homogenous fibroblast-like morphology,the shapes of cells were polygonal,and spindle;90.60% ADSCs were G0/G1 phase cells;the positive rates of cellular surface antigenes were CD44 99.88%,CD105 95.00%,CD90 99.74%,CD49d 89.56%,CD34 3.04%,CD106 0.85%,CD45 0.26%;after induction,intracellular lipid droplets could be observed and present jacinth by Oil Red stain in 14 days.Conclusions:ADSCs are easily isolated and cultured in vitro,and have the potential to differentiate into adipogenic cells.ADSCs can be as the seed cells for the treatment of POP.
