摘 要:目的筛选出对HBV有抑制作用的反义寡核苷酸(AS-ONs),为HBV的预防和治疗提供基础。方法合成5条与HBV互补的反义寡核苷酸,转染HepG2.2.15细胞,运用荧光定量PCR和酶联免疫法测定反义寡核苷酸对细胞培养液中HBVDNA和HBsAg、HBeAg的抑制作用。结果寡核苷酸序列Ⅰ(TAC CTC TTG TAG TGT AGT CCT)在HepG2.2.15细胞能抑制HB-sAg、HBeAg、HBV的DNA表达。结论靶向C基因区AS-ONs能够剂量依赖性地抑制HBV的复制,可能发展成为新型抗HBV药物。Objective To study screening of antisense oligonucleotides targeting HBV and to provide great help for the prevention and treatment of HBV.Methods 5 antisense oligonucleotides targeting HBV were synthesized and transfected to HepG2.2.15 cells.The detection of antisense oligonucleotides inhibitory effects on HBV DNA.HBeAg and HBsAg were performed using Real time PCR and ELASI.Results The replication of HBV DNA and expression of HBeAg and HBsAg reduced by the antisense oligonucleotides in the HepG2.2.15 cells.Conclusion The antisense oligonucleotides targeting HBV has does-depend inhibitory effects on HBV DNA replication and reduces the expression of HBeAg and HBsAg.It maybe a new potential drug targets for anti-HBV.
