CD_(137)在人外周血T细胞的表达及其介导信号对T细胞增殖的影响

摘　　要：为研究ＣＤ１３７在正常人静止状态和活化Ｔ细胞上的表达特点及ＣＤ１３７单抗协同ＰＨＡ 刺激Ｔ细胞增殖作用。利用流式细胞术测定ＣＤ１３７的表达，利用３Ｈ－ＴｄＲ掺入法测定Ｔ细胞增殖。结果显示：静止Ｔ细胞上ＣＤ１３７表达率为（０．６５±０．０５）％ ，经ＰＨＡ 刺激后２４、４８、７２ｈ，Ｔ 细胞上ＣＤ１３７表达率分别为（８．１６±１．２８）％ 、（１２．３９±２．１５）％ 、（２１．６±４．３６）％ 。ＣＤ＋４ Ｔ细胞和ＣＤ＋８ Ｔ细胞均可表达ＣＤ１３７，且ＣＤ＋４ Ｔ细胞的表达率高于ＣＤ＋８ Ｔ细胞。单独用ＣＤ１３７单抗对Ｔ细胞的刺激指数（ＳＩ）为１．０４，单独用ＰＨＡ 刺激指数为２．３１，不同浓度ＣＤ１３７单抗（１、５、１０）ｍ ｇ／Ｌ和ＰＨＡ 共同作用刺激指数分别为２．８３、４．２５、５．７４。ＣＤ１３７ＭｃＡｂ（１０ｍ ｇ／Ｌ）＋ ＰＨＡ 组刺激指数比其他各组均明显增高（Ｐ＜ ０．０５），并随ＣＤ１３７ＭｃＡｂ 浓度增高对Ｔ 细胞的增殖逐渐增高。表明静止Ｔ细胞上不表达ＣＤ１３７，Ｔ细胞活化后，ＣＤ１３７表达量逐渐增加。ＣＤ１３７单抗可显著增加ＰＨＡ对Ｔ细胞的增殖作用，而在无ＰＨＡ 时。To study the expression of CD 137 on still T cell and active T cell in normal human and the effect of CD 137 McAb on PHA stimulated T cell proliferation, the expression of CD 137 was measured by flow cytometry (FCM), T cell proliferation was measured by [ 3H]thymidine incorporation assay. The result indicated that the expression rate of CD 137 were(8.16±1.28)%,(12.39±2.15)%,(21.6±4.36)% respectively after T cell is stimulated by PHA 24h, 48h,72h.CD 137 is expressed on both CD + 4T cell and CD + 8T cell, but the expression rate on CD + 4T cell is higher than that on CD + 8 T cell. Stimulation index (SI) of CD 137 McAb stimulated T cell proliferation was 1.04, SI of PHA stimulated was 2.31. SIs of different concentration (1mg/l,5mg/l,10mg/l) CD 137 McAb+PHA stimulated were 2.83, 4.25, 5.74 repectively, SI of CD 137 McAb(10mg/L)+PHA stimulated is higher than that on the other groups (P<0.05),when the concentratiion of CD 137 McAb is increased followed by increasing of the proliferation of T cell. It indicated that CD 137 was not expressed on still T cell, after T cell is activated, the expression rate of CD 137 gradually increased. CD 137 McAb can stimulate T cell proliferation in the presence of PHA, but CD 137 McAb has no effect in the absence of PHA.
