摘  要:为了鉴定脱氢表雄酮抗体的特异性及使用效价,观察脱氢表雄酮在人早期胎盘绒毛的细胞定位。方法:免疫组织化学ABC法。结果:脱氢表雄酮抗体稀释度为1:1000-1:2000时,胎盘切片仍显示较强的脱氢表雄酮免疫反应阳性,抗体经过量的脱氢表雄酮一BSA偶联复合物吸收后,再进行孵育时,胎盘切片呈阴性反应,提示该脱氢表雄酮抗体有较高的使用效价及特异性。人胎盘绒毛两层滋养层细胞和基质细胞均呈脱氢表雄酮免疫反应性,阳性反应物质主要分布在胞质,偶尔也在胞核内,提示人胎盘绒毛以上三种细胞可能能合成脱氢表雄酮,脱氢表雄酮对胎盘发育及妊娠维持可能有重要调节功能。To study sensitivity and specificity of rabbit anti dehydroepiandrosterone(DHEA) antibody and localization of DHEA in human placenta villi. Methods: The immunohistochemical ABC method was used in the experiment. Results: The placenta villi still appeared strong immunoreactivity in l: 1000--1: 2000 dilution of DHEA antibody and the substitution and absorption tests were negative, suggesting that the DHEA antibody had high sensitivity and specificity. The cytotrophoblast, syncytiotrophoblast and stroma cells all showed strong DHEA immunoreactivity. The positive substance was distributed in the cytoplasm and occasionally in the nuclei, suggesting that these cells may produce DHEA, and DHEA plays an important role in placenta developing and pregnancy maintaining.
