前凝血状态下激素性股骨头缺血坏死血管壁骨代谢相关基因差异表达的实验研究

摘　　要：目的:运用基因芯片技术,对前凝血状态下激素影响的血管壁中的骨代谢相关基因进行筛选,发现相关基因的差异表达情况,为进一步探讨前凝血状态下激素导致股骨头缺血坏死的发病机理提供新的思路。方法:选健康中国大白兔25只,分为对照组、内毒素组、模型组,利用大肠杆菌内毒素及激素诱导兔典型前凝血状态下激素性股骨头缺血坏死模型,取主要供应股骨头血供的旋股内侧动、静脉,提取组织总RNA,与含有82个兔基因的寡核苷酸基因芯片杂交,洗涤,染色和扫描,分析检测的数据。结果:在检测的82个基因中,下调的基因动脉有51个,静脉有56个,其中表达差异显著的基因动脉有7个,静脉有23个。结论:(1)激素在导致股骨头发生缺血坏死的过程中降低或增高了某些血管壁中主要的骨代谢相关基因的转录。进而影响了其在血管壁中的生物活性。所有这些可能都会影响到股骨头局部的骨代谢,从而加重或导致了股骨头坏死。(2)利用基因芯片大通量筛选的技术,在短时间内可以迅速准确的分析激素影响的血管壁中众多骨代谢相关因子及其受体的基因表达情况,为进一步研究激素性股骨头坏死机制提供了有效的手段。Objective： To study the pathogenesis of steroid-induced osteonecrosis of the femoral head,using Microarray to find differentially expressed genes from blood vessel wall,find the major regulated gene and anlysis.To provide the new ideas and further discussion the pathogenesis of steroid damaged the blood vessel caused the femoral head necrosis finally.Methods：twenty-five white rabbits were divided into three groups randomly,control group,LPS group and model group.Establish the model of steroid-induced avascular necrosis of femoral head and examine them with electron microscope.Get the medial femoral circumflex artery and vein.Total RNA of the tissues was extracted and hybridized with the genes on the gengchip containing 122 genes.Results：In 82 genes there were 7 genes differentially expressed in artery and 23 in vein.we find these genes correlated with angiogenesis,ossification.Conclusion：（1）In the process of steroid-induced avascular necrosis of femoral head,steroid down-regulated angiogenic growth factors and ossific related factors.So,the biological activity of these fractors were effected in the blood vessel wall.Vascular regeneration and bone regeneration were influenced caused new osteonecrosis.Down-regulation of angiogenic growth factors was the important pathological basis of steroid-induced osteonecrosis.（2）It is concluded that many growth factors and receptors could be analyzed in a short period by using geng chips,which provided a powerful method for further studies on steroid-induced osteonecrosis of the femoral head.
