摘  要:目的:采用人脐带间充质干细胞(human umbilical cord mesenchymal stem cells,hUCMSCs)与蚕丝蛋白生物支架构建组织工程化脂肪组织,研究不同移植部位对组织工程化脂肪存活的影响。方法:将人脐带间充质干细胞接种于蚕丝蛋白支架上,体外成脂诱导培养1~2周,扫描电镜观察细胞在支架上的生长及黏附情况;同时,将细胞-支架复合物分别移植于6只Wistar雌性大鼠背部皮下和股二头肌内,观察其成脂及存活情况。结果:移植8周、12周后,组织冰冻切片油红O染色、HE染色及扫描电镜显示,实验组移植复合物内有脂肪细胞生成,背部移植复合物表面皮肤结合紧密,与底部肌肉组织结合疏松,体积较移植前增大,支架材料随移植时间延长逐渐降解;股二头肌内移植复合物移植8周后,与肌肉组织结合紧密,但体积较移植前明显减小,支架材料降解明显,到移植12周后移植处不见移植物存在。空白对照组移植物内无脂肪细胞生成,移植物体积较植入前减小,股二头肌内移植物减小更明显;股二头肌内移植物移植12周后,移植处不见移植物存在。结论:蚕丝蛋白生物支架具有良好的三维空间结构和生物降解性,人脐带间充质干细胞接种于蚕丝蛋白生物支架可用于组织工程化脂肪组织的研究;不同移植部位对该组织工程化脂肪存活有一定的影响,而背部皮下为良好的移植位点。Objective To construct the tissue engineering adipose in vivo with human umbilical cord mesenchymal stem cells(hUCMSCs) and silk fibroin scaffold,and research the effect of different transplantation spots on the survival of tissue engineering adipose.Methods hUCMSCs were seeded in silk fibroin scaffold and cultured one to two weeks in vitro.The adhesion and growth of cells were observed with scanning electron microscope.Meantime,the cell-scaffold complexes were allografted in subcutaneous on back and biceps femoris of 6 female Wistar rats.Results The new formed adipose tissue was observed on 8 and 12 weeks later.In experimental group,the grafting complexes had adipocytes,and the grafting complexes in back combine with the subcutaneous closely and with the bottom muscle loosely;the size was larger than that before transplantation.AT the 8th week,the grafting complexes in biceps femoris combined with the muscle closely,but the size was obviously smaller than that before,the scaffols degraded obvously.In control group,the grafting has no adipocytes and the size was getting smaller,especially the grafting in biceps femoris.After grafted 12 weeks,the grafting in biceps femoris was not exsisted in the transplantation spot.Conclusions The silk fibroin scaffold is a new type of scaffold with 3D space structure and good biodegradation.hUCMSCs could be used in the research of tissue engineering adipose.Different transplantation spots have an effect on the survival of tissue engineering adipose partly,and the subcutaneous of back is an ideal transplantation spot.
