摘 要:为宁强矮马、哈萨克马的分子系统学地位和生长机制的研究提供分子水平的科学依据,对宁强矮马、哈萨克马和其他动物生长激素受体基因外显子10序列进行分析,通过PCR扩增宁强矮马和哈萨克马的生长激素受体基因外显子10,并进行测序,用DNAMAN软件对不同动物的生长激素受体(GHR)基因外显子10序列进行比对分析,并以生长激素受体(GHR)基因外显子10序列为基本数据,对不同物种进行系统树构建。结果表明,所研究的个体间有1个SNP位点,以家马生长激素受体基因外显子108序列(AF39287)为标准,宁强矮马539位点缺失1个碱基A,哈萨克马该位点由A→G,这些变异导致氨基酸发生改变,可能影响其生物学功能。通过系统树构建发现,在生长激素受体(GHR)上,所选动物的亲缘关系与生物进化树一致。The sequences of grown hormone receptor(GHR) exon 10 of Ningqiang pony,Kazakh horse,and other animals were studied by PCR amplification,gene sequencing of Ningqiang pony and Kazakh horse,software DNAMAN was used for comparative analyze of the sequences of GHR exon10 in different animals,and consequently constructed the phylogenic tree based on GHR gene exon 10 sequence in different animals to provide the scientific basis in molecular level for the study on molecular systematic status and growth mechanisms of Ningqiang pony and Kazakh horse.The results showed that there was one SNP locus in the coding region.There was a base deletion A in 539 locus of Ningqiang pony and a transition of A to G in Kazakh horse in this locus compared with the standard sequence of AF39287 in horse,these variation caused the changes of amino acid,which might affect its biological function.The construction of phylogenic tree showed that the genetic relationship of studied animals was identical with the biological cladogram in terms of the sequence of GHR.
