人肺腺癌细胞株A549和GLC-82的计算机图像色度学定量分析

摘　　要：目的从色度学角度定量分析人肺腺癌细胞株A549和GLC-82的相似性和差异性,为更好地认识、分析、理解及比较这两株细胞的有关研究结果奠定基础。方法用计算机图像分析技术分别测试A549和GLC-82细胞蜡块切片之HE染色和细胞甩片之巴氏染色的色度学参数,包括细胞浆的红、绿、蓝三基色(Rp、Gp、Bp)及其三基色系数(rp、gp、bp),核的红、绿、蓝三基色(Rn、Gn、Bn)及其三基色系数(rn、gn、bn)。结果色度学参数测试结果显示:除Gp外,A549和GLC-82细胞甩片巴氏染色细胞及胞核三基色参数Rp、Rn、Gn、Bp、Bn及其三基色系数rp、rn、gp、gn、bp、bn差异均具有显著性(P<0.01);A549细胞的Rp、Rn、rp、rn均高于GLC-82细胞,GLC-82细胞的Gp、Gn、Bp、Bn、gp、gn、bp、bn均高于A549细胞。GLC-82细胞蜡块切片HE染色细胞及胞核之Rp、Rn、Gp、Gn、Bp、Bn、gp、gn、bn均高于A549细胞(P<0.05),而A549细胞的rp、rn、bp均高于GLC-82细胞(P<0.01)。结论人肺腺癌细胞株A549和GLC-82细胞蜡块切片HE染色之色度学参数Rp、Rn、Gp、Gn、Bp、Bn、rp、rn、gp、gn、bp、bn差异具有显著性;两者细胞甩片巴氏染色之色度学参数Rp、Rn、Gn、Bp、Bn、rp、rn、gp、gn、bp、bn差异同样具有显著性,这些差异有助于这两株细胞的甄别。Objective To analyze the differences and the similarities of the chromatic characteristics between human lung adenocarcinoma cell lines A549 and GLC-82.Methods Cytological samples of A549 and GLC-82 cells were prepared from paraffin sections stained with hematoxylin and eosin,and by sedimentation centrifugation and papanicolaou staining.Computer image analysis of the cytological samples was performed to determine the chromatic parameters,including three primary colors of cytoplasm（Rp,Gp,Bp） and their coefficient（rp,gp,bp）,three primary colors of cell nuclei（Rn,Gn,Gn） and their coefficient（rn,gn,bn）.Results The measurement results of chromatic parameters showed that in the cytology by sedimentation centrifugation and papanicolaou staining,the values of Rp,Rn,Gn,Bp,Bn,rp,rn,gp,gn,bp and bn had significant difference between A549 and GLC-82 cells（P0.01）;the values of Rp,Rn,rp,rn in A549 cells were higher than those in GLC-82 cells,while the values of Gn,Bp,Bn,gp,gn,bp,bn of GLC-82 cells were higher than those of A549 cells.On the HE-stained paraffin sections,the values of Rp,Rn,Gp,Gn,Bp,Bn,gp,gn and bn of GLC-82 cells were higher than those of A549 cells（P0.05）,while the values of rp,rn,bp of A549 cells were higher than those of GLC-82 cells（P0.01）.Conclusions There are significant differences between the chromatic parameters of Rp,Rn,Gp,Gn,Bp,Bn,rp,rn,gp,gn,bp and bn of A549 and GLC-82 cells,not only in HE-stained paraffin sections,but also in sedimentation centrifugation cytological slides by Papanicolaou staining（except for Gp）.These differences could be used for the identification between A549 and GLC-82 cells.
