摘  要:目的:研究紫杉醇对Burkitt淋巴瘤细胞株Daudi细胞凋亡的影响。方法:用原子力显微镜(AFM)分析细胞膜超微结构的改变,CCK8法检测Daudi细胞增殖活性,流式细胞术(FCM)测定凋亡率。结果:AFM显示紫杉醇可引起细胞形貌的改变,正常Daudi细胞呈圆形,表面较光滑;紫杉醇处理后,Daudi细胞坍塌,细胞表面粗糙、凹凸不平。紫杉醇对Daudi细胞的增殖具有抑制作用,且呈时间浓度依赖性。FCM检测示Daudi细胞凋亡率各实验组均明显高于对照组(P<0.05),呈浓度依赖性。结论:紫杉醇可改变细胞膜超微结构,抑制Daudi细胞的增殖,促进其凋亡。Objective To investigate the effects of paclitaxel on the apoptosis of Burkitt lymphoma Daudi cell lines. Methods The membrane uhrastrueture of Daudi cells was analyzed by AFM, the proliferation was checked up with CCK8, the apoptosis was investigated by flow cytometer quantitatively. Results Paelitaxel could change tile morphology of Daudi cells to varying degrees, normal Daudi cell was globular and the surface was smooth. After being treated by paclitaxel, the Daudi cell eollapsed, cell surface was roughness and was full of bumps and boles. The proliferation of Daudi cells could be inhibited by paelitaxel in a lime-and-concentrationdependent manner. As compared with that in control group, the apoptosis rate of Daudi cells were significantly increased in all the experiment groups (P 〈 0.05) in a concentration- dependent manner. Conclusion Paclitaxel can change the membrane ultrastrueture of Daudi cells, inhibit cell growth, and induce apoptosis.
