摘  要:蛋白阵列(Protein array)也叫蛋白芯片,是生物芯片(Biochip)的一种,主要利用抗原/抗体可特异性结合原理制备而成。按阵列样点大小,蛋白阵列又可分为微阵列(Microarray)和宏阵列(Mac-roarray)两类。Acidovorax avenae subsp.citrulli(Aac) is a seedborne pathogen harmed cucurbitaceae plants se-riously.In this research,a protein macroarray was developed to detect the pathogen rapidly using chemiluminescence.On nitrocellulose membrane,a macroarray was fabricated and it showed satisfactory results in the detectability.The macroarray could detect Aac as low as 2.3×105 cfu/mL without apparently crossing reaction with other seedborne pathogens of cucurbitaceae crops,and it showed a same detectability with ELISA for seed samples.This method also had a good specificity and repeatability.Moreover,60 seed samples were used to test the practicability of this detection method.The results showed that the detection system had a high consistent(60/60) with ELISA assay.This system had identical detectability with only 1/12 capture antibody and 1/8 detection time of ELISA assay and the operation was very convenient.After assembling with a hybri-dization box,it could be generalized to grass roots workstation.Meanwhile,this work provided a basis of simultaneous detection to multiple plant pathogens using protein macroarray.
