摘  要:以通过溶胶-凝胶技术制备的SiO2/Nafion杂化膜固定辣根过氧化物酶,以杂化膜中的Nafion固定的亚甲基蓝为辣根过氧化物酶和玻碳电极间的电子传递介体,制成了电流型单酶过氧化氢生物传感器。在此基础上通过固定双酶(辣根过氧化物酶-葡萄糖氧化酶)制成了葡萄糖生物传感器。探讨了杂化膜的制备条件、生物传感器的性能和工作电位、pH值、温度、干扰物质等对生物传感器的影响。单酶生物传感器线性响应范围为1.0×10-6~1.6×10-4mol/L,检测限为6.0×10-7mol/L(S/N=3),达到95%稳态响应电流用时少于15s。双酶葡萄糖生物传感器线性响应范围为7.8×10-6~2.4×10-3mol/L。检测限为4.2×10-6mol/L(S/N=3),达到95%稳态响应电流用时少于25s。An amperometric biosensor highly sensitive to H2O2 is constructed by immobilizing horseradish peroxidase(HRP) and Methylene Blue into the sol-gel-derived SiO2/Nafion hybrid film.The presence of Nafion in the sol-gel-derived hybrid film not only prevents the cracking of the pure sol-gel-derived SiO2 film but also immobilizes positively charged organic dyes such as methylene blue(MB).The immobilized MB acts as electron transfer mediator between horseradish peroxidase and glass carbon electrode.Similarly,an amperometric biosensor highly sensitive to glucose is constructed by immobilizing HRP and glucose oxidase(GOD) into the hybrid film of sol-gel-derived SiO2 and Nafion.Preparation conditions of the hybrid membrane and Characteristics of the biosensors and dependence of the biosensors on applied potential,solution pH,temperature and interferents are discussed in details.The single enzyme biosensor shows a wide linear response to hydrogen peroxide in the range of 1.0×10-6 ~ 1.6×10-4 mol/L with a detection limit of 6.0×10-7 mol/L(S/N=3) and reaches 95% of steady-state current less than 15s.While the dual enzyme biosensor shows a linear response to glucose in the range of 7.8×10-6 ~ 2.4×10-6 mol/L with a detection limit of 4.2×10-6 mol/L(S/N=3) and reaches 95% of steady-state current less than 25s.
