吲达帕胺对脂多糖诱导的心肌细胞损伤的保护作用

摘  要:目的探讨吲达帕胺对脂多糖(LPS)诱导的心肌细胞损伤的保护作用。方法取SD新生大鼠45只,分离心肌细胞培养,在培养的第4天随机分为3组:对照组(n=15,行常规培养)、LPS组(n=15,于心肌细胞中加入1 mg.L-1的LPS培养6 h)、吲达帕胺+LPS组(n=15,于心肌细胞中加入吲达帕胺0.01 mg.L-1处理12 h后,加入1 mg.L-1的LPS培养6 h)。分别观察3组心肌细胞搏动频率、存活率和培养液中乳酸脱氢酶(LDH)活性。结果 1)心肌细胞搏动频率:LPS组比对照组明显减低(P<0.01);吲达帕胺+LPS组比LPS组显著加快(P<0.01),比对照组稍慢,但差异无统计学意义(P>0.05)。2)细胞存活率:LPS组比对照组明显降低(P<0.01);吲达帕胺+LPS组比LPS组明显增高(P<0.01),与对照组相比差异无统计学意义(P>0.05)。3)LDH活性:LPS组比对照组明显升高(P<0.01);吲达帕胺+LPS组比LPS组明显降低差异(P<0.01),与对照组相比虽有升高但差异无统计学意义(P>0.05)。结论从细胞水平证实吲达帕胺对心肌细胞LPS损伤具有保护作用。Objective To investigate the protective effects of Indapamide on the cultured cardiocytes injuried by lipopolysaccharide(LPS).Methods Cultured cardiocytes of 45 neonatal SD rats were randomly divided into three groups at the 4th day(15 rats every group):control group(conventional culture),LPS group(adding LPS 1 mg·L^-1 culture 6 h),Indapamide+LPS group(adding Indapapamide 0.01 mg·L^-1 culture 12 h,LPS 1 mg·L^-1 culture 6 h).At the end of LPS experiment,cardiocytes beating rate,cell viability and the activity of LDH in culture were measured.Results The beating rate of LPS group was significantly slower versus control group(P0.01),Indapamide+LPS group was significantly faster versus LPS group(P0.01),and no difference exists between Indapamide+LPS and control group(P0.05).The cell viability of LPS group was significantly decreased versus control group(P0.01),Indapamide+LPS group was significantly higher versus LPS group(P0.01),and no diffrerence exists between Indapamide+LPS and control group(P0.05).The LDH activity of LPS group was significantly increased versus control group(P0.01),Indapamide+LPS group was significantly lower compared by LPS group(P0.01),and no diffrerence exists between Indapamide+LPS and control group(P0.05).Conclusion Indapamide could alleviate cardiac damage produced by LPS on the cellular.
