摘 要:目的探讨纳米铜粉对去卵巢大鼠骨量丢失的预防作用及其机制。方法 48只去卵巢大鼠随机分为正常对照组、模型对照组、骨疏康组(1.8g/kg)与纳米铜粉0.84、2.53和7.58mg/kg组,观察纳米铜粉对大鼠骨密度(BMD)、断裂载荷及股骨湿重、长度、直径和体积的影响,检测成骨样及成纤维细胞与用药大鼠血清共同培养后的增生率。结果与模型对照组相比,纳米铜粉2.53和7.58mg/kg组骨质疏松大鼠BMD、断裂载荷增加(P<0.05或P<0.01),纳米铜粉7.58mg/kg组大鼠股骨体积与股骨长度增高(P<0.05)。纳米铜粉7.58mg/kg组成骨样及成纤维细胞增生率可分别达到39.4%和64.7%。结论纳米铜粉可增加骨质疏松大鼠股骨BMD、断裂载荷与股骨体积和长度,经纳米铜粉治疗的骨质疏松大鼠血清可促进成骨样和成纤维细胞增生。Objective To investigate the preventive effect and mechanism of nanometer copper on the bone loss in ovariectomized rats.Methods A total of 48 ovariectomized rats were randomly divided into control group,model group, Gushukang group(1.8 g/kg)and nanometer copper 0.84,2.53 and 7.58 mg/kg group.The effects of nanometer copper on bone mineral density(BMD),fracture load,wet weight,length,diameter and volume of femur were observed.Osteoblastlike cells and fiber cells were used as osteoblast model and the cell proliferations were measured.Results Compared with the model group,the BMD and fracture load of nanometer copper 2.53 and 7.58 mg/kg groups increased(P0.05 or P0.01),and the wet weight and volume of femur in 7.58 mg/kg nanometer copper group increased(P0.05).The proliferations of osteoblast-like and fiber cell were 39.4%and 64.7%in nanometer copper 7.58 mg/kg group.Conclusion Nanometer copper can increase the BMD,fracture load,wet weight and volume of femur.The serum of osteoporosis model rats treated with nanometer copper could promote the osteoblast-like and fiber cell proliferation.
