摘 要:为了应用生物信息学分析绵羊FGF5基因编码蛋白的结构与抗原表位,预测该蛋白的免疫原性。根据牛的FGF5基因cDNA保守区序列设计引物对绵羊FGF5基因cDNA进行扩增,将扩增片段进行克隆和测序,并与牛的FGF5基因序列进行同源性比较,确定扩增的片段为绵羊FGF5基因片段。利用蛋白分析专家系统(ExPASy)提供的ProtParam、SOSUI、MotifScan、SWISS-MODEL等生物信息学在线分析程序,结合TMH-MM2.0、SignalP3.0、TargetP 1.1和PSORTⅡprediction、Gene Runner和DNAMAN等生物信息学软件,分析、预测FGF5蛋白的理化性质、可溶性、翻译后修饰位点、亲(疏)水性、跨膜区、信号肽序列、蛋白细胞定位区域、二级结构、抗原表位等。结果显示该蛋白由270个氨基酸组成,分子式C1315H2085N389O378S6,分子质量单位为29.5847 ku,理论等电点为10.59,波长280 nm时的吸光度(A)值为0.864,半衰期为30 h,22个翻译后修饰位点、多个亲水性高的区域、2个跨膜区域、11个潜在抗原表位,为可溶性和分泌型蛋白,具有免疫原性,可据此制备多克隆抗体。To predict the immunogenicity of the FGF5 protein coded for by the FGF5 from sheep and characterize its structure and epitopes from a bioinformatics approach,the cDNA of FGF5 gene was cloned from sheep by using PCR with a pair of primers designed according to the conserved cDNA sequence of cow homologous FGF5 gene.The physico-chemical properties,solubility,surface accessibility,post-translational modification sites,hydrophilicity/hydrophobicity,secondary structure,specific epitopes,and 3D structure of FGF5 protein were analyzed and predicted by using the ExPASy bioinformatics tools ProtParam,SOSUI,Emini,MotifScan,HNN,and SWISS-MODEL in combination with bioinformatics software(TMH-MM2.0,SignalP 3.0,TargetP 1.1和PSORTⅡprediction,Gene Runner,DNAMAN,etc.).Results showed that FGF5 protein consisted of 270 amino acids and had the following characteristics:molecular formula:C1315H2085N389O378S6;molecular mass:29.5847 ku;theoretical isoelectric point:10.59;absorbance(A):0.864(280 nm);half-life:30 h.22 post-translational modification sites,numerous highly hydrophilic regions,and 11 potential epitopes expressed as a soluble protein.The FGF5 protein coded for by the FGF5 gene may be made polyclonal antibody because of its soluble protein expression and immunogenicity.
