摘 要:目的运用大孔吸附树脂分离纯化桔梗总皂苷,为桔梗总皂苷的生产提供参考。方法采用单因素试验结合正交试验法,以桔梗总皂苷含量、保留率为指标,对桔梗总皂苷的纯化工艺进行优选。结果最佳条件为选用D101型大孔树脂,上样浓度0.5g生药材/ml,径高比1:9,吸附流速为2BV/h,吸附2h后,以6BV水洗脱,再分别以5BV25%、70%乙醇洗脱,解析流速为3BV/h,收集70%乙醇洗脱液,纯化后桔梗总皂苷的含量达到56.32%,保留率为74.86%。结论 D101树脂对桔梗总皂苷具有较好的吸附和解析性能,纯化工艺简单可行、重复性好,具有良好的产业化前景。Objective Purification of platycodins by macroreticular resin to provide evidence for their industrialized production. Methods A reasonable purifying process was established by a combination of single-factor experiments and orthogonal designs, using the content and retention rate of platycodins as the evaluation makers. Results The optimum conditions were as follows: the type of macroreticular resin was D101, the concentration of loading sample was 0.5g/ml, the ratio of diameter-length was 1:9, the absorbing velocity of flow was 2BV/h, After an absorption period of 2 hours, the column was eluted with 5BV water and then with 5BV 25%, 70%ethanol at a flow rate of 3BV/h, respectively, collected the 70% ethanol eluent, after purification of platycodins by macroreticular resin, the content of platycodins was 56.32% and the retention rate was 74.86%. Conclusion D101 macroreticular resin had good properties of absorption and resolution for platycodins, the process was simple and reproducible, with good prospects for industrial.
