摘  要:通过优化预处理和检测方法,采用Waters C18反相色谱柱,以磷酸缓冲盐和乙腈为流动相,外标法测定GA3的回收率为78.42%,变异系数±3.9,保留时间稳定,首次建立了毛竹种子中植物内源激素GA3含量的测定方法。运用此法测定毛竹种子萌发过程中GA3含量的变化,结果表明,从浸种到出苗时GA3的含量在2.56~14.75μg.g-1,吸胀阶段含量变化不明显,而浸种36 h后含量大幅升高,在72 h和即将萌出胚根时达到峰值,随着毛竹种子的萌发进程,GA3含量呈总体上升趋势。The measurement of endogenous GA3 contents in moso bamboo seed was set up for the first time by optimizing pretreatment and determination method,which was carried out with external standard method on a reverse phase Waters C18 column using phosphoric acid buffer and acetonitrile as mobile phase.The results showed that recovery rate was 78.42%,the RSD was ± 0.039,and retention time was stable.Changes within the germination of the moso bamboo seed in the content of gibberellins were studied during sprouting.GA3 level in-creased from 2.56 μg·g-1 to 14.75 μg·g-1 during sprouting process.GA3 levels were low at the beginning of the soaking,and then increased greatly after 36 h after soaking to the highest peak at 72 h until radicle appeared.The levels of GA3 were higher in radicle than that in embryo.
