摘  要:目的:探讨地黄饮子含药脑脊液对β-淀粉样蛋白(Aβ25-35)所致PC12细胞损伤的β-淀粉样前体蛋白(APP)mRNA的表达及地黄饮子对其的干预作用。方法:运用中药脑脊液药理学方法把PC12细胞分为空白组,模型组,西药对照组,地黄饮子脑脊液低,中,高剂量组6组,分别加入正常培养液,正常脑脊液及含药脑脊液,应用实时定量PCR法检测受损PC12细胞β-淀粉样前体蛋白mRNA表达(用2-ΔΔC t表示,Ct为阈循环值,ΔΔCt=ΔCt各干预组-ΔCt空白组,ΔCt=CtAPP-CtGAPDH)。结果:地黄饮子能明显降低PC12细胞受β-淀粉样蛋白刺激时APP的过度反应,降低APP mRNA的表达,与模型组比较,差异有统计学意义(P<0.01)。结论:地黄饮子脑脊液可以抑制β-淀粉样蛋白损伤对PC12细胞的损伤。Objective:To discuss the cerebrospinal fluid with ancient prescriptions Dihuangyinzi which has a protective function by the APP mRNA on PC12 cells injured by β-amyloid protein(Aβ25-35).Method: We divide PC12 cells into 6 groups-that is,the blank group,the model group,the control group of western medicine,and the cerebrospinal fluid groups with low-dose,medium-dose and high-dose of Dihuangyinzi with pharmacology method of cerebrospinal fluid containing Chinese medicine;and add normal culture medium,the normal cerebrospinal fluid and cerebrospinal fluid containing drugs respectively;APPmRNA expression in PC12 cells by real-time quantitative reverse transcription-PCR(expressed as 2-ΔΔCt,Ct as cycle threshold,ΔCt=Ct APP-Ct GAPDH,ΔΔCt=ΔCt invervention groups-ΔCt blank controls group).Results: Decoction of Dihuangyinzi can significantly reduce the excessive reaction with the stimulation by the Aβ on PC12 cells and the Protein expression of APP with the model group;the difference has a statistics significance(P0.01).Conclusion:Cerebrospinal fluid with Dihuangyinzi can inhibit the PC12 cells injury by Aβ.
